摘 要:目的了解不同切应力下肝窦内皮细胞分泌肝细胞生长因子(HGF)、白细胞介素-6(IL-6)、一氧化氮(NO)及一氧化氮合成酶(NOS)的分泌量,探讨切应力对原代培养的大鼠肝窦内皮细胞分泌功能的影响。方法构建血流动力学模型,实验分为对照组(切应力为0dyn/cm2)和实验组2组,实验组又按切应力不同分为12、24和48dyn/cm23个亚组。每组分别在4、8、12及24h抽取1ml培养基,利用试剂盒检测培养基中HGF、IL-6、NO和NOS浓度。结果在不同切应力下,肝窦内皮细胞分泌HGF、IL-6、NO和NOS各不相同,在同一时相随着切应力的升高,HGF、IL-6、NO和NOS的分泌量也升高,实验组明显高于对照组(P<0.01),且48dyn/cm2组明显高于另两个亚组(P<0.01);实验组作用8、12、24h时,HGF、IL-6、No及NOS分泌量明显高于作用4h时。结论体外实验证实,切应力升高时肝窦内皮细胞分泌HGF、IL-6、NO和NOS的量明显增加,提示部分肝切除术后门静脉压力急剧升高能促使肝窦内皮细胞活化,分泌大量促肝细胞再生的细胞因子和介质。Objective To observe concentration claanges of substances including growth factor (HGF),interleukin-6(IL-6) ,nitric oxide(NO), nitric oxide synthase(NOS) that were secreted by sinusoidal endothelial cells (SECs) in different shear stress and study the effects of shear stress on the secretions of SECs. Methods To establish a hemodynamics equipment and to set up a way to separate and culture SEC. The rats were divided into two groups: the control group (shear stress was 0 dyn/cm^2) and study group. Study group was divided into three sub-groups according to shear stress (12, 24 and 48 dyn/cm^2). Sinusoidal endothelial cell culture mediums were examined in different shear stress and phases. Results HGF, IL-6, NO and NOS secreted by sinusoidal endothelial cells in different shear stress were distinctive and had positive correlation with shear stress and action time. Conclusion In vitro, the secretion of HGF,IL-6,NO and NOS of sinusoidal endothelial cells are increased along with shear stress rising, which suggests that the rising pressure in portal vein after hepatectomy may result in SECs activated. Activated SECs then secret many cytokines and medias to trigger the regeneration of liver.
