广西壮族与汉族人群IL-1β基因5号外显子+3953C/T多态性的研究

摘　　要：目的观察白细胞介素-1β(IL-1β)基因5号外显子+3953C/T多态性在中国广西壮、汉两民族中的分布。方法应用聚合酶链反应-限制性片段长度多态性技术检测155名壮族人和195名汉族人的白细胞介素-1β基因5号外显子+3953C/T多态性,计算其基因型和等位基因频率,并结合文献与高加索人、非洲白人进行了不同种族间的分析比较。结果在壮族人中CC、CT基因型分别为94.8%、5.2%;在汉族人中CC、CT基因型分别为93.3%、6.7%。两组IL-1β基因型的分布频率差异无显著性(P>0.05)。二者与高加索人、非洲白人比较,发现IL-1β基因型分布及等位基因频率有显著差异(P<0.01)。结论IL-1β基因5号外显子+3953C/T多态性在广西壮、汉两族分布无差异,但与高加索人、非洲白人分布有差异。Objective To study the distribution of the fifth exon region ＋ 3953C/T polymorphism of interleukin-1β gene in the chinese Zhuang and Hart ethnic groups in Guangxi province. Methods The fifth exon region ＋ 3953C/T polymorphism of interleukin-1β gene was detected by polymerase chain reaction-restriction fragment length polymorphism technique in healthy Zhuangs（ n = 155） and Hans（ n = 195）. Results The frequencies of CC and CT genotypes were found to be 94.8% and 5.2% in Zhuangs, 93.3% and 6.7% in Hans respectively. There was no significant difference in the frequency distribution of interleukin-1β genotype between Zhuangs and Hans （ P 〉 0.05 ）, but the polymorphism of IL-1β （ ＋ 3953 ） in Zhuangs and Hans was significantly different, compared with Caucasinans and African white populations （ P 〈 0.01 ）. Conclusion There was not significant difference of the fifth exon region ＋ 3953C/T polymorphism of interleukin-1β gene between Zhuangs and Hans, but there were obvious differences in Zhuangs and Hans compared with Caucasinans and African whites.
