摘  要:在转入含反义蜡质基因T-DNA水稻后代中获得白化突变体。本试验从遗传分析、电镜观察和电泳分析对该白化突变体进行研究。结果显示,该白化突变体是由于在转基因组培操作过程中由核基因突变产生的,能够稳定遗传,属于单基因位点隐性突变;超微结构观察显示,白化苗具有前质体,但没有叶绿体;SDS-PAGE分析,三叶期白化苗比绿苗缺少3条条带,分子量分别约为112.36 KD、18.08 KD和9.65 KD。本研究为进一步开展导致该白化突变体的功能基因分析奠定了基础。An albino mutant was obtained from transgenic rice progenies into which an anti-waxy gene had been introduced. The albino mutant was researched on inheritance analysis, ultrastructure observation and SDS-PAGE. The results showed that the albino mutant was due to nuclear gene mutation in tissue culture during the transformation, being stable hereditary. The albino character was controlled by a recessive single gene, and it belongs to a permanent albino type, Uhrastructure observation revealed that there were some proplastids, but no chloroplast in the albino-seedlings, SDS-PAGE analysis indicated that the albino-seedling lacked three polypeptide bands ( 112.36 KD, 18.08 KD and 9.65 KD ) compared with the green-seedling. The present study laid a foundation of further researches on the function gene resulting in this albino mutation.
