高脂高糖诱导的2型糖尿病大鼠肾脏细胞凋亡的分析

摘　　要：目的研究分析高脂高糖诱导的2型糖尿病大鼠肾脏细胞凋亡的变化。方法用高糖高脂饲料(10·0%猪油,20·0%蔗糖,5·0%胆固醇,65·0%常规饲料)喂养SD大鼠诱发胰岛素抵抗,然后用小剂量链脲佐菌素(STZ)腹腔注射,诱发2型糖尿病,用TdT介导的脱氧核苷酸切口末端标记法(TdT-mediateddUTPnickendlabel-ing,TUNEL)检测肾脏细胞凋亡,并与常规饮食大鼠(对照组)相比较。结果2型糖尿病大鼠肾脏细胞的凋亡数显著多于对照组(P<0·0001)。结论本研究为2型糖尿病大鼠肾脏细胞的凋亡提供证据,提示凋亡在肾脏进行性损伤中起重要作用,细胞凋亡与糖尿病肾病的发生密切相关。Objective To study the apoptosis in the type 2 diabetes meUitus rats kidney. Methods SD rats were fed with the diets enriched with sucrose （20%） and lard （10%） and cholesterol （5%） to induce insulin resistance.Type 2 diabetes meUitus was developed by intmperitoneal injection in these rats with 25mg/Kg streptozocin （STZ） . Rats Kidney cells apoptosis was detected by in situ TUNEL staining. Results The number of apoptosis cells in type 2 diabetes mellitus rats kidney was higher than teat in control （ P〈0.0001 ） . Conclusion The present study provides evidence for apoptosis in type 2 diabetes mellitus rats kidney. It suggests a role for apoptosis in the gradual loss of renal mass and an intimate relationship between apoptesis and diabetic nephropathy.
