摘　　要：用进口和国产医用硅橡胶制成一根型双烯高诺酮(Gestodene)埋植剂,长度为40、20和10mm(合药75、37.5和18.75mg),内填充Gestodene和硅橡胶混炼的药芯,外包裹硅橡胶释药管,两端粘合剂封口.体外释药量测试用紫外分光光度法.三种剂量的埋植剂273天体外释放相当稳定.大鼠10个月药效试验无一例怀孕,大鼠体内平均日释放量为60.7、29.0和13.4μg.经10个月动物试验后取出埋植剂分析其残留药量,10个月内耗药量占总药量的24、23和21%.A single rod implant containing Gestodene was prepared from medical silicone rubber which was manufactured at home and abroad and was 40, 20 and 10 mm in length (containing 75, 37. 5 and 18. 75 mg of Gestodene respectively). The implant was filled with Gestodene in the core of silicone rubber and sealed at both ends with a special silicone rubber medical adhesive. In vitro release of Gestodene was determined by UV spectrophotometry.Implants containing the various doses of the drug all released Gestodene rather constantly during the period of 273 days. No pregnancy occurred in rats during 10 months. The average daily release of Gestodene in vivo was 60. 7, 29. 0 and 13. 4 ?g respectively. The drug residual in the implants after a 10 months experiment showed the 24, 23 and 21% of the initial load of drug were delivered in the animal body.
