摘 要:使用2×107PFU乙脑病毒野毒株GSS抗原preM-E-NS1-NS2a的非复制型重组痘苗病毒NTV-JEV免疫3周龄BALB/c小鼠,小鼠体内乙脑病毒特异性抗体滴度1∶71,IgG2a/IgG1为1.5,中和抗体滴度为1∶8,免疫组小鼠产生特异性补体介导的细胞杀伤作用。以10 LD50乙脑P3株病毒对小鼠进行了脑内攻击,NTV-JEV免疫组小鼠存活率为100%,与乙脑减毒活疫苗SA14-14-2免疫组存活率相同。100 LD50P3株病毒攻击后NTV-JEV免疫组存活率28.6%,与SA14-14-2组免疫效果无明显差别。存活小鼠体内乙脑病毒特异性抗体升高为1∶179,IgG2a/IgG1比值为1.9,中和抗体大于1∶16。以30 LD50GSS株病毒进行攻击后,NTV-JEV免疫组和SA14-14-2组保护率均为8.3%。上述结果表明,NTV-JEV与乙脑减毒活疫苗SA14-14-2株具有相近的保护力,NTV-JEV不仅可以保护小鼠抵抗同株病毒GSS的攻击,而且可以抵抗异株病毒P3的攻击。Four groups of 3-week-old BalB/c mice were immunized twice by intramuscular (i. m. ) injection with 2 × 10^7 PFU/mouse of non-replicating vaccinia recombinant NTV-JEV which expresses the preM-E-NS1-NS2a antigens of Japanese encephalitis virus(JEV). Three weeks after the second immunization, the mice produced JEV-specific antibody with total antibody titer of 1 : 71, IgG2a/IgG1 ratio of 1.5 and neutralizing antibody of 1 : 8. Antibody-dependent complement-mediated cytotoxic activity was detected only in NTV-JEV immunized mice but not in mice immunized with attenuated live vaccine SA14-14-2 . After challenged with 10 LD50 JEV P3 strain, 100 % of the mice immunized with either NTV-JEV or attenuated live vaccine SA14-14-2 were survived. After challenged with 100 LD50 P3 strain, only 28.6% of the mice in NTV-JEV immunized group were survived, that was similar to that in attenuated live vaccine SA14-14-2 immunized group. JEV-specific antibody titer in the survived mice increased to 1 : 179, the ratio of IgG2a/IgG1 was 1.9, and the neutralizing antibody titer exceeded 1 : 16. When challenged with 30 LDs0 of GSS strain, both the groups immunized with NTV-JEV and attenuated vaccine SA14-14-2 had the same survival ratio of 8.3 %. All the data above showed that NTV- JEV has the similar protective efficacy with that of the attenuated live vaccine SA14-14-2. NTV-JEV induced protective immunity against challenge not only with the homologous strain GSS but also with the heterologous strain P3.
