摘  要:①目的探讨丙氧鸟苷(GCV)对亚致死量照射H2半相合小鼠CD34+细胞移植的影响。②方法从亲代雄性供鼠骨髓分离CD34+细胞,同时取脾脏T细胞转染TK基因。雌性F1受鼠60Co亚致死量照射7.0Gy后随机分为4组,第1组输入1×105CD34+细胞,其他3组输入1×105CD34+细胞和5×105TK+T细胞。第3、4组分别于移植后0和7d给予GCV治疗,然后观察各组移植物抗宿主病(GVHD)发生情况和生存情况。③结果单纯输入CD34+细胞组小鼠未能植活,但自体造血恢复。输入CD34+和TK+T细胞组小鼠出现急性GVHD,表现为体质量下降;自小鼠外周血扩增出Y染色体片段。输入CD34+和TK+T细胞后应用GCV组小鼠未发生急性GVHD;0d应用GCV组80%小鼠未扩增出Y染色体片段;7d应用GCV组80%小鼠扩增出Y染色体片段。7d应用GCV组植入率与未应用GCV组相比差异无显著性(χ2=0.89,P>0.05);7d应用GCV组植入率高于0d应用组(χ2=2.64,P<0.05)。④结论移植后1周应用GCV对CD34+细胞植入影响较小。Objective To explore the impact of ganciclovir(GCV) administration on the engraftment of CD34^+ progenitors in sublethally-irradiated H-2 haploidentical transplantation. Methods CD34^+ progenitors were separated from male parental donors, bone marrows, and T lymphocytes gained from their spleens and transduced by retroviral vectors encoding TK gene. Female F1 recipient mice were sublethally irradiated (7.0 Gy) and randomly divided into four groups. Recipient mice in group 1 only received 1×10^5 CD34^+ progenitors and those in other three groups received 1 × 10^5 CD34^+ progenitors and 5 × 10^5 TK^+ T lymphocytes. GCV was administered on 0 or 7th posttransplant day in groups 3 and 4, respectively. Acute GVHD and survival were observed, Results Engraftment did not occur in recipient mice who only received CD34^+ progenitors, but autologous hematopoiesis recovered, The mice received CD34^+ progenitors and TK^+ T lymphocytes manifested acute graft-versus-host disease with the decrease of body weight, Chromatosome Y segments were enlarged in peripheral lymphocytes. No aGVHD symptoms appeared in the mice received CD34^+ progenitors and TK^+ T lymphocytes and administered GCV, Chromatosome Y segments were not enlarged in 80% of the mice who were administered GCV on 0 d, while chromatosome Y segments were enlarged in 80% of the mice who were administered GCV on 7th day, There was no difference in engrafting ratios between the group that received CD34^+ progenitors and TK^+ T lymphocytes without GCV and that administered GCV on 7th day (χ^2 =0, 89.P〉0. 05), Engrafting ratio of the group administered GCV on 7th day was higher than that of the group administered GCV on 0 d (χ^2 = 2. 64, P〈0. 05), Conclusion There is less influence on the engraftment of CD34^+ progenitors when GCV is administered one week after the transplantation.
