摘 要:目的观察局灶性脑缺血再灌注大鼠学习记忆与海马齿状回神经元型一氧化氮合酶(nNOS)表达的关系。方法大脑中动脉线栓法制作大鼠局灶性脑缺血再灌注模型,5溴脱氧尿核苷(BrdU)标记分裂增殖细胞,应用Y型电迷宫监测大鼠学习记忆能力,免疫组化单标和双标技术检测大鼠海马齿状回BrdU阳性细胞数和nNOS的表达。结果局灶性脑缺血再灌注对学习记忆能力损害明显,但随着再灌注时间的延长学习记忆能力有较好恢复,再灌注28d恢复明显。海马齿状回BrdU阳性细胞增加,14d达高峰;再灌注后7dnNOS表达增强,28d达高峰,BrdUnNOS双标细胞在14d达高峰。结论局灶性脑缺血再灌注可增强大鼠海马齿状回细胞的增殖能力,增殖细胞nNOS的表达对大鼠学习记忆能力恢复有促进作用。Objective To observe the relationship between the recovery of learning and memory and the expression of nNOS positive cells in the dentate gyrus (DG) of hippocampus in adult rats after focal cerebral ischemia. Methods In middle cerebral artery occlusion (MCAO) models, bromodeoxyuridine (BrdU) was injected to label proliferative cells. The Y-type electronical maze was used to monitor the recovery of learning and memory of every adult rats. Then BrdU positive cells and nNOS positive cells in DG were observed with immunohistochemistry. Resuits In MCAO rats, the ability of learning and memory were injured severely, but it recovered better after reperfusion 28 d. BrdU positive cells in DG obviously increased and arrived at the peak on reperfusion 14 d. nNOS expression in DG also increased after reperfusion 7 d and arrived at the peak on reperfusion 28 d. BrdU/nNOS double positive cells increased and arrived at the peak on reperfusion 14 d. Conclusions The ability of proliferation of cells in DG can be improved after focal cerebral ischemia, and some of them express nNOS. It may be related to the recovery of learning and memory after focal cerebral ischemia.
