摘  要:目的通过对我国青海田鼠与布氏田鼠鼠疫菌生物学特性的比较,了解两种菌株的遗传特点及其亲缘关系。方法以生化、毒力因子、毒力、PstⅠ敏感性、质粒、外膜蛋白,重复片断扩增多态性指纹图谱,脉冲场电泳图谱等方法进行比较。结果青海田鼠与布氏田鼠鼠疫菌株的主要生化性状均为鼠李糖(+)、麦芽糖(+)、密二糖(+)、甘油(+)、阿胶糖(-)脱氮(-);具有FI+、VW+、Pgm+、PstⅠ+;对小白鼠表现为毒力强,青海田鼠型鼠疫菌对豚鼠的毒力高于布氏田鼠型鼠疫菌;两种菌株均对鼠疫菌(包括自身)产生的PstⅠ敏感;它们均携带(6、45、65)×106相对分子质量(Mr)3种质粒,外膜蛋白图谱均缺失32kd条带;重复片断扩增多态性指纹图表现出二者有近缘关系,脉冲场凝胶电泳图谱一致。结论青海田鼠与布氏田鼠鼠疫菌的生物学特性相似,有较近的亲缘关系。Objective To compare the biological features of Yersiniapestis(Y. pestis) Isolated from Microtus Fuscus with that from Microtus Brandti, the genetic features and relationship of the two types of Y, pestis are found. Methods The biochemical features, virulence factors, virulence, Pst Ⅰ sensitivity, plasmids, outer membrane proteins, repeated amplified fragment polymorphisms and pulsed field gel electrophoresis spectrum were studied. Results The main biochemical features of the two types of Y. pestis were Rha(+) ,Mal(+) ,Mel(+), Gly(+), Ara(-) ,Den(-), FI^+, VW^+, pgm^+, PstⅠ^+. The virulence of the two types of Y. pestis were powerful to mice, that ofMicrotus Fuscus was more powerful than that of Microtus braadti. The two types of Y. pestis were sensitive to Pst Ⅰ , and they carried three kinds of plasmids of 6 × 10^6, 45× 10^6 and 65 × 10^6. Their outer membrane proteins spectrums lacked of the 32 × 10^3 band. The two types had a similar repeated amplified fragment polymorphism spectrum and an identical pulsed field gel electrophoresis spectrum. Conclusion The two types of Y. pestis have similar biological and genetic features, which revealed their close affinity.
