摘 要:目的:研究APA微囊猪胰岛的体外体内功能。方法:用胶原酶灌注消化分离猪胰岛,用葡聚糖(dextran)梯度密度离心纯化胰岛,用海藻酸钠聚赖氨酸海藻酸钠(APA)包埋胰岛,用静态培养和灌流刺激实验评价猪胰岛体外功能,植入用注射链脲佐菌素(STZ)制作的糖尿病大鼠腹腔评价体内功能。结果:经分离、纯化和包埋后的微囊猪胰岛无论体外静态培养或灌流均能释放胰岛素,并能对培养液中的葡萄糖浓度发生应答。植入糖尿病大鼠腹腔,能降低血糖并分泌胰岛素,维持时间达6个月。结论:APA微囊猪胰岛无论体外体内均能保持较好的功能,对治疗糖尿病动物模型有较好疗效。Objective: The purpose of this study was to evaluate the functions of APA microencapsulated porcine islets. Methods: Islets were isolated from porcine using collagenase perfusion technique, purified by dextran histopaque gradients and encapsulated with alginate-polylysine-alginate (APA) microcapsules prepared by the droplet generation technique. APA microencapsulated porcine islets were stimulated with glucouse when cultured in vitro and perfused with glucose of different concentrations. Diabetes Mellitus (DM) animal models were induced by intraperitoneal injection of STZ and then received intraperitoneal transplantation of microencapsulated islets. Results: After isolation, purification and encapsulation, APA microencapsulated porcine islets expressed the ability to secrete insulin both in vitro culture and in perfusion condition, and the islets showed the ability of responsing to the glucose concentration in culture medium. The levels of blood sugar of DM rats were decreased for 6 month after intraperitoneal translpantation of microencapsulated islets. Conclusions: APA microencapsulated porcine islets maintained good function both in vitro and in vivo, and had better therapeutic effect on diabetes animal model.
