摘 要:目的:了解活血化瘀中药三七和丹参对视网膜新生血管形成的小鼠血管内皮细胞生长因子的影响。方法:实验于2003-05/2003-06在广州中医药大学中药学院完成。C57BL/6J幼小鼠40只,随机分为正常对照组、模型对照组、三七组、丹参组,每组10只。建立氧诱导的血管增生性视网膜病变幼鼠模型,模型对照组、三七组、丹参组动物均置于密闭容器内,容器内氧气分压控制为75%;正常对照组在正常空气环境下饲养。5d后将动物取出置于正常空气环境下饲养,三七组每天腹腔注射血栓通注射液(三七总皂苷35g/L)1次,丹参组每天腹腔注射丹参注射液1次,按10mL/kg体质量给药,正常对照组及模型对照组每天腹腔注射等容积生理盐水1次。5d后所有动物自眼球取血,采用双抗体夹心法测定血清血管内皮细胞生长因子含量。结果:实验小鼠40只全部进入结果分析。血管内皮细胞生长因子含量:丹参组低于模型对照组[(15.65±2.51),(19.34±4.88)ng/L,P<0.05];三七组和正常对照组低于模型对照组[(12.87±3.26),(9.28±1.26)ng/L,P<0.01]。结论:活血化瘀药物三七和丹参能减少血管增生性视网膜病变小鼠的血管内皮生长因子含量,可通过改善局部缺氧环境或影响各种新生血管生长因子而阻止视网膜新生血管形成。主题词:视网膜新生血管化/中药疗法;三七/药理学;丹参/药物作用;AIM: To understand the effects of the huoxue huayu traditional Chinese medicine,sanchi and dan-shen root on the vascular endothelial growth factors formed by retina newly vessel in mice. METHODS: The experiment was done in the College of Traditional Chinese Medicine, Guangzhou University of Traditional Chinese Medicine from May 2003 to June 2003. Forty C57BL/6J young mice were selected and divided randomly into normal control group, model control group, sanchi group and dan-shen root group with 10 mice in each group. The young mice models with vessel hyperplasia retinopathy induced by oxygen were made. The mice in the model control group, the sanchi group and the dan-shen root were all put into the close container; the pressure of the oxygen in the container was 75%; The mice in the normal control group were raised in the normal air condition. Five days later, the mice were gotten out of the container to raise in the normal air condition. The mice in the sanchi group were treated with xueshuan tong parenteral solution (sanchi saponin 35 g/L) by intraperitoneal injection once every day. The mice in dan-shen root group were treated with dan-shen root parenteral solution once every day by intraperitoneal injection, administration according to 10 mL/kg body mass. The mice in the normal control group and the model control group were treated with the equal bulk of saline by intraperitoneal injection once every day. Five days later blood of the eyes was gained from all the animals. The content of growth factors of serum blood vessel endothelial cells were detected by double antibody with filling method. RESULTS: Forty experimental mice were all involved in the result analysis. The content of the growth factors of the vessel endothelial cells: It was lower in the dan-shen root group than that in the model control group [( 15.65±2.51 ), (19.34±4.88) ng/L, P 〈 0.05]; It was lower in the danchi group and the normal control group than that in the model control group [(12.87±3.26), (9.28±1.26�
