摘  要:目的体外观察ConA活化淋巴细胞来源的DNA(ALDDNA)对BALB/c小鼠巨噬细胞株RAW264.7的刺激作用。方法将ALDDNA及未活化淋巴细胞来源的DNA(UnALDDNA)分别与BALB/c小鼠来源的巨噬细胞株RAW264.7孵育48h,然后以四甲基偶氮唑蓝(MTT)法检测细胞的增殖情况,流式细胞仪测定细胞表面分子的表达,ELISA检测白细胞介素6(IL6)、白细胞介素12(IL12)和肿瘤坏死因子α(TNFα)的分泌情况。结果ALDDNA在体外能够明显刺激细胞发生增殖,上调细胞表面主要组织相容性复合体(MHC)II类分子以及共刺激分子CD40、CD80和CD86的表达,并能够诱导细胞分泌IL6、IL12和TNFα。结论ALDDNA对RAW264.7细胞具有免疫刺激作用。Purpose To observe the immunostimulatory effect of Con A-activated lymphocytes derived DNA(ALD-DNA) on RAW264.7 in vitro. Methods Macrophage cell line RAW264.7 from BALB/c mice was incubated 48 hours with ALD-DNA and unactivated lymphocytes derived DNA (UnALD-DNA), respectively. The proliferation of RAW264.7 was determined by MTT colorimetric assay. The cell-surface expression of MHC class Ⅱ and costimulatory molecules CD40, CD80, CD86 was analyzed by FACS. The production of IL-6, IL-12 and TNF-α by RAW264.7 was assessed by ELISA. Results ALD-DNA could trigger proliferation of RAW264. 7, up-regulate MHC class Ⅱ and costimulatory molecules prensent on RAW264.7 in vitro,and stimulate secretion of IL-6, IL-12 and TNF-α. Conclusions ALD-DNA has the immunostimulatory effect on RAW264.7.
