摘 要:目的探讨尼美舒利联合阿霉素能否增强对肝癌细胞生长的抑制。方法采用MTT法测定尼美舒利、阿霉素及尼美舒利联合阿霉素对SMMC7721、HepG2细胞体外增殖的抑制率,统计学及金正均法q值判断两药的相互作用。结果不同浓度的尼美舒利和阿霉素可抑制SMMC7721、HepG2的增殖,并呈剂量依赖性,尼美舒利和阿霉素联合使用抑制作用明显增强,尼美舒利25、50、100μmol/L分别与阿霉素0.15625、0.3175、0.625、1.25μmol/L合用时,q值均>1.15,呈现协同作用。统计学处理结果均显示两种药物间存在交互作用。结论一定浓度的尼美舒利在体外能增强阿霉素对肝癌细胞生长的抑制;尼美舒利与阿霉素抗肝细胞癌作用具有协同效应。Objective To study the inhibition effects of combination of nimesulide and doxorubicin on proliferation of hepatocellular carcinoma cells(HCC). Methods Hepatocellular carcinoma cell SMMC7721 and HepG2 were treated with nimesulide and doxorubicin. Cell survival rate was determined by MTT assay. The interaction of nimesulide and doxorubicin was estimated by q value provided by Jin Zhengjun's method. Results The combination of nimesulide and doxorubicin significantly inhibited the growth of SMMC7721 and HepG2. The synergistic effects were seen in combination of nimesulide 25,50, 100 μmol/L and doxorubicin 0. 156 25, 0. 317 5,0. 625, 1.25 μmol/ L respectively. All q values 〉 1. 15. Statistics results showed that there was an interaction between two drugs. Conclusion Nimesulide can enhance the antiproliferative effect of doxorubicin and the combination of two drugs presents synergistic effect.
