摘 要:目的观察粉防己碱对兔实验性动脉粥样硬化形成的影响。方法健康♂新西兰幼年兔60只,随机分为3组,对照组以普通饲料喂养,高脂模型组和粉防己碱组以高脂饲料喂养,粉防己碱组同时静注粉防己碱5mg.kg-1.d-1。观察各组实验前、喂养6和12 wk时的体重和血脂的变化,及12 wk结束时主动脉壁和心肌小动脉的变化。结果各组相同时间的体重、对照组实验前、喂养6和12 wk后的血脂水平均差异无显著意义(P>0.05);模型组12 wk TG、TC、LDL、HDL分别为(1.97±0.83)、(19.22±3.10)(、17.50±2.64)(、0.25±0.09)mmol.L-1,与对照组[分别为(0.86±0.93)、(0.82±0.16)、(1.77±1.78)、(0.40±0.14)mmol.L-1]相比差异有显著性意义(P<0.01),与粉防己碱组[分别为(2.35±0.28)、(20.39±2.94)、(18.52±1.80)、(0.25±0.11)mmol.L-1]相比差异无显著性意义(P>0.05)。12 wk末,对照组主动脉内膜光滑,表面无纤维斑块形成,模型组动脉壁内膜表面有较多大小不同呈黄色粥样的纤维斑块形成,粉防己碱组内膜表面有奶油色或乳白色变化,但无凸出于表面的纤维斑块。结论粉防己碱不能降血脂,但可抑制动脉粥样硬化的形成和进展。AIM To study the effect of tetradrine(Tet) on atherosclerosis in high-fat rabbits. METHODS Sixty healthy New Zealand male juvenile rabbits, were divided randomly into three groups, control group fed with common diet, model group with high-fat diet and Tet group with high-fat diet plus Tet, which were giver by venous injection (5 mg · kg^-1·d^-1) for 12 wk. Every animals were weighed and determined plasma lipid level before experiment, at the 6th week and at the 12 th week respectively. At the end of the 12 th week, all rabbits were killed, the aorta walls and small cornary artery were observed by naked eyes or microscope. RESULTS The difference in weight among three groups at the same time was nonsignificant(P 〉0.05); at the end of the 12th week, TG, TC, LDL, HDL of the model group was ( 1.97 ± 0.83), ( 19.22 ± 3.10), ( 17.50 ± 2.64) and (0.25 ± 0.09) mmol· L^- 1 respectively, compared with the control group [ (0.86 ± 0.93), (0.82 ± 0.16), (1.77 ± 1.78) and (0.40 ± 0.14)mol· L^-1 respectively ], the difference was significant (P 〈 0.01), but compared with the Tet group[(2.35 ± 0.28), (20.39 ± 2.94), (18.52 ± 1.80), (0.25 ± 0.11) mmol·L^-l respectively] ,the difference was nosignificant (P 〉 0.05). The artery walls in control group were smooth whithout fibrous plaque, while a veriety of shape yellow fibrous plaque in model group and only the fatty streak in Tet group were found. CONCLUSION The results suggestes that Tet may affect the formation of fibrous plaque, i. e. it's anti-atherosclerosis effect, although without reducing high plasma lipid level in high-fat diet rabbits.
