三苯氧胺对小鼠内脏器官影响的病理学研究

摘  要:目的观察三苯氧胺(TAM)对小鼠内脏器官的形态影响。方法以不同剂量的TAM(0.02mg.kg-1.d-1、0.2mg.kg-1.d-1、2mg.kg-1.d-1)分别连续灌胃染毒小鼠35天,于末次染毒称取体重后处死小鼠,取出心、肝、肾等器官,称重,光镜下观察病理形态变化。结果高剂量组与阴性对照组比较,肝/体系数差异有显著性(P<0.05),部分肝细胞可见脂肪变性。心、肾脏体系数与阴性对照组比较差异无显著性,光镜下无形态异常。结论部分肝细胞脂肪变性可能与长期服用TAM有一定的关系。Objective To observe the effects of tamoxifen(TAM)on the internal organ of mice. Methods TAM was administrated to mice at different doses(0. 02mg· kg^-1·d^-1 ,0. 2mg· kg^-1· d^-1 , 2mg · kg^-1· d^-1 )by gavage once a day for successive 35 days. The mice were weighted and sacrificed on the 35th day. The hearts, livers and kidneys were enucleated and weighted. The pathomorphologic observation were made by light microscope . Results The ratio of liver/body had significant difference between 2mg· kg^-1· d^-1 groups and negative control group (P〈0. 05). The liver cells with fatty degeneration were observed in 2mg· kg^-1· d^-1 and 0. 2mg· kg^-1· d^-1 TAM groups. The ratio of heart and kidney to body had no significant difference with those in negative control group. Conclusion Hepatic steatosis may occurred after lasting taking TAM.
