足阳明经穴针刺血清对家兔离体胃窦平滑肌细胞内钙离子浓度影响的实验研究

摘　　要：目的:观察足阳明经穴针刺血清对家兔离体胃窦平滑肌细胞内钙离子(Ca2+)浓度的影响。方法:将45只家兔随机分为5组各9只。生理盐水组,耳缘静脉滴注0.9%NaCl溶液;阿托品模型组,耳缘静脉滴注以生理盐水配制的2.5mg/100ml的阿托品;足阳明经穴组,造模并电针双侧足三里、天枢、梁门、四白穴;足少阳经穴组,造模并电针双侧阳陵泉、带脉、日月、阳白穴;足太阳经穴组,造模并电针双侧承筋、背下点、背上点、攒竹穴。分组处理后,颈动脉采血制备血清。另取正常家兔10只,制备正常兔胃窦平滑肌细胞悬液。取各组1:2稀释的血清与同体积的正常兔胃窦平滑肌细胞悬液作用,以荧光分光光度计测定细胞内Ca2+浓度。结果:阿托品模型组Ca2+浓度低于生理盐水组,差异有显著性意义(P<0.05);足阳明经穴组Ca2+浓度明显高于生理盐水组、阿托品模型组、足少阳经穴组、足太阳经穴组,差异均有非常显著性意义(P<0.01);足少阳经穴组与足太阳经穴组比较,差异无显著性意义(P>0.05)。结论:足阳明经穴针刺血清使离体胃窦平滑肌细胞内Ca2+浓度升高具有经脉的相对特异性。Objective： To explore the influence of serum after acupuncture of Foot Yangming Meridian on intracellular concentration in smooth muscle of isolated rabbit gastric antrum. Methods： 45 rabbits were allocated randomly to normal saline group （NSG）, atropine model group （AMG）, Foot Yangming Points acupuncture group （YMAG）, Foot Shaoyang Points acupuncture group（SYAG） and Foot Taiyang Points group （TYAG）. NSG： 0. 9% NaCl solution was injected i. v. by drips through otomarginal Vein. AMG： 2.5mg/100ml atropine （prepared with normal saline） was injected i.v. drips through otomarginal vein. YAMG： electroacupuncture （EA） of bilateral Zusanli, Tianshu, Liangmen and Sibai Points after modelling. SYAG： EA bilateral Yanglingquan, Daimai, Riyue and Yangbai points after modelling. TYAG： EA of bilateral Chengjin, Beixiadian, Beishangdian, and Zanzhu points. After treatment, blood was collected from carotid artery, and acupuncture serum（AS） was prepared. Other 10 normal rabbits were used to prepare smooth muscle cellular suspension of gastric antrum. The intracellular Ca^2＋ concentration in 1 ： 2 diluted AS and same volume of smooth muscle cellular suspension was determined by fluoro spectrophoto meter. Results： The influence on intracellular Ca^2＋ are as follows： AMG was lower than NSG （ P 〈 0. 05） ; YMAG was higher than NSG, AMG, SYAG and TYAG （ P 〈 0. 01 ）; no significant difference between TYAG and SYAG （P 〈 0.05）, indicating serum after acupuncture Foot Yangming Meridian can increase the intracellular Ca^2＋ Concentration, but that after acupuncture Foot shaoyang and Foot Taiyang acupoints cannot. Conclusion： The serum after acupuncture of Foot Yangming Meridian can increase intracellular Ca^2＋ concentration in smooth muscle of isolated rabbit gastric antrum with relative specificity.
