摘 要:目的探讨3种聚丙烯酰胺凝胶提纯电泳在蛋白质提纯中的应用。方法用普通聚丙烯酰胺凝胶提纯电泳(PAGE)、等电聚焦提纯电泳(IEF)和等速提纯电泳(ITP)对样品进行分离。结果3种提纯电泳均可分离蛋白质。PAGE的区带分辨率低,加样量不高;IEF的区带分辨率最高,可分离同种蛋白的亚成分,加样量最小;ITP区带分辨率较高,可将样品分成单一成分,加样量最大。结论低离子强度提纯电泳是可行的,分离效果令人满意。Objective To evaluate three kinds of polyacrylamide gel electrophoresis in purification of protein. Methods The protein was purified by common polyacrylamide gel electrophoresis (PAGE) , isoelectric focusing electrophoresis (1EF) and isotachophoresis (1TP). Results The protein could be isolated by three kinds of purification electrophoresis. The resolving power of PAGE was low and the load of sample was low. The resolving power of 1EF was the highest and the load of sample was the lowest. The resolving power of ITP was high and the load of sample was the highest. Conclusions The purification electrophoresis can be operated in low ionic strength. The result is satisfying.
