摘　　要：目的探讨不同时段哮喘大鼠肺组织CD44和α-平滑肌肌动蛋白(α-SMA)mRNA的表达,了解其在哮喘发病中的作用机制。方法以卵蛋白致敏制备大鼠哮喘模型.分别在激发后1、2周取肺组织.RT-PCR检测肺组织CD44与α-SMAmRNA的表达,收集肺泡灌洗液(BALF)行细胞分类计数.肺组织石蜡切片行HE染色。结果哮喘大鼠BALF中炎性细胞明显增多(P<0.05).CD44与α-SMA在正常大鼠肺组织少量表达,在激发哮喘后1周肺组织CD44与α-SMAmRNA表达均显著升高(P均<0.05).2周后升高更明显(P<0.01)。CD44和α-SMA的表达呈正相关.CD44mRNA水平与BALF中淋巴细胞和嗜酸性粒细胞的百分比呈显著正相关。结论肺组织CD44与α-SMA在转录水平的过量表达与哮喘的发生发展密切相关,二者存在相互作用.并在早期开始参与哮喘的发病。CD44与嗜酸性粒细胞、淋巴细胞向肺部炎症组织趋化浸润密切相关。Objective To explore the expressions of xmooth muscle actin （SMA） -α and CD44 mRNA in the lungs of rats with asthma in different stages. Methods The asthmatic rat model was established by repeated exposing the animal to aerosolized ovalbumin （OVA）. After being sensitized. the rat was subjected to laboratory test. ,Semi - quantitatively reverse transcription - polymerase chain reaction （RT- PCR） was used to analyze CD44 and α- SMA nRNA of lung tissue. Differential leukocytes（ neutrophils, eosinophils, and lymphocytes）counts in bronchoalveolar lavage fluid （BALF）were performed after Wright staining. Results The expressions of CD44 and α -SMA nRNA increasted after OVA challenge 1 week （P〈0.05） and increased significantly after OVA challenge 2 weeks （P〈0.01 ）. There was significant increase in lymphocytes and eosinophils in BALF in model group compared with normal group（ P〈 0.05）. A positive correlation between CD44 and α-SMA was demonstrated. CD44 expression was significantly correlated with the percentage of lymphocytes and eosinophilic granulocytes in BALF. Conclusions In the rats with asthma, cell adhesion molecule CD44 and α-SMA have important roles in the cascade of inflammation and the development. CD44 has an important function on promoting leukocyte recruitment to the lungs.
