摘  要:为探讨家蚕丝腺细胞合成分泌蛋白质过程中有关基因的表达调控机理,采用蛋白质双向电泳和图像分析技术,研究了家蚕4眠期和5龄期后部丝腺细胞的蛋白质组成。在4眠家蚕的后部丝腺细胞中检测到425个蛋白斑点,5龄期随着生长发育的进程,可检测到新的特异蛋白斑点,说明不同的发育时期,后部丝腺细胞的蛋白组成是不一致的。5龄前、中期新增加的特异蛋白斑点可能与丝素蛋白的合成、分泌有关,而5龄后期新增加的特异蛋白斑点可能与丝腺组织的形态变化、细胞的分解凋亡有关。Changes of protein forms from posterior silkgland of silkworm Bombyx mori between larvae of 4^th molting and the 5^th instar stage were analyzed by using 2D-PAGE and image analysis technique for inquiring into regulation mechnism of relative gene expression during synthesis and secretion proteins in silkgrand cell. The 425 protein spots were detected in posterior silkgland of 4^th molting larva. New special protein spots could be detected along with growth of the silkworm, which revaled that the protein forms in posterior silkgland cell is different at different stage. The special protein spots appeared during prophase and metaphase of 5^th instar stage may associated with the synthesis and secretion of fibroin protein, while the special protein spots appeared in anaphase of 5^th instar stage may associated with the changes of configuration of silkgland tissue and silkgland cell apoptosis.
