摘 要:采用蛋白质双向电泳和图像分析技术,对温敏性家蚕品种K05在不同温湿度催青条件下不同发育阶段胚胎的蛋白质变化进行研究,发现蛋白斑点1、7、8、10、13、15、17、18、19、20和25的缺失、蛋白斑点11、12、16、21、22、23和24含量的降低与高温干燥条件下催青时胚胎的不孵化有关;而蛋白斑点2、3、5、6和9的特异表达,以及蛋白斑点4和14含量的增加,与高温多湿催青条件下胚胎正常发育孵化有关。由此推测:温敏性家蚕品种在高温干燥催青条件下不孵化的原因可能是由于一系列基因的被关闭或表达量下降所造成;而在高温多湿条件下催青时胚胎能够正常发育孵化,则可能是由于一些特异基因被激活,致使温敏性家蚕品种的生理代谢得到了补偿。The proteins extracted from different stages embryo of temperature sensitive silkworm breed K05, incubated under different temperature and humidity, were researched by two-dimensional polyacryamide gel electrophoresis (2D-PAGE) and image analysis system. It is discovered that the missing of protein spots of 1, 7,8,10,13,15,17,18,19,20 and 25 as well as the decreased expression of protein spots of 11,12,16,21,22, 23 and 24 are related with non-hatching when incubated under high temperature and dry condition, while the distinctive expression of protein spots of 2,3,5,6 and 9 as well as the increased expression of protein spots of 4 and 14 may contribute to the normal hatching when incubated under high temperature and wet condition, from which we may speculate as follows: (1)the non-hatching when incubated under high temperature and dry condition may attributed to that a series of genes were turned off or down; (2) while the embryo developed smoothly when incubated under high temperature and wet condition may attributed to a series of distinctive gene's activation, which compensated physiologic metabolism of temperature sensitive silkworm breed.
