欧达胃克对大白鼠长期毒性的观察

摘　　要：应用大白鼠连续ig给药4周,对欧达胃克的长期用药毒性进行观察,结果,低剂量组(350mg·kg^(-1);相当于临床量的2.3倍)和中剂量组(700mg·kg^(-1),4.5倍量)与对照组比较各项指标无显著差异。高剂量组(1232.0mg·kg^(-1),8倍量)用药2周活动逐渐减少,饮食量下降,体重减轻。用药3周高剂量组有30％大鼠站立行走不便,出现肌张力障碍。测大鼠坐骨神经复合动作电位,表现抑制现象,兴奋阈降低。高剂量组BUN有可逆性升高。实验结束时有3只大鼠死于呼吸抑制(死亡率未超过20％)。The rats were given drugs intra-gaster (ig) for four weeks to observe lie long-trem toxicictis 01 auduweike. And the results-showed that there were no differences among lowest dose group (350 mg/kg equal 2.3 times clinical tose), middle dose group (700mg/kg 4.5 times dose ) and control group 1232 mg/kg, 8 times dose).The weight, activity and alimentary quantity of rats ail-reduced. For three weeks, about 30% of the rats in this group could not stand and walk freely and appear myodystony. The compound action proten-icl of aervas ischiadicus expressed inhibtion state, and the threshold of ex-itability declined. The blood urea nitrogen of high dose group rose reversi-bilitily. At the end of the experiment, 3 rats died of respiratory depression (mortaly20%)
