摘  要:目的观察大鼠连续腹腔注射(ip)丹参葡萄糖注射液(salvia miltiorrhiza glucose injection,SGI)产生的毒性反应与靶器官损害的可逆性。方法通过血液学、血液生物化学和病理组织学检查,观察SGI对大鼠的毒性反应及其程度。结果连续ip SGI 26周,动物全部存活。大剂量组动物进食量减少,体重增加较慢,肝功能AST升高,凝血时间延长,对肝脏有一定损伤,停药后均减轻或消失。中、小剂量组动物未见明显毒性反应。结论ip SGI,大剂量[12.5 g/(kg.d)]可抑制大鼠体重增长,(7/22)动物肝细胞呈浊肿、变性;中剂量[5.0 g/(kg.d)](4/22)动物肝细胞有轻度浊肿,肝损有一定可逆性。提示主要靶器官是肝脏,停药后各剂量组肝脏均未见明显病损。Objective To investigate the toxicity and the impaired reversibility on target organ after administering ip salvia miltiorrhiza glucose injection(SGI) in rats. Methods The toxicity reaction and impaired degree after administering SGI were observed by detecting indexes of hematology, hematological biochemics and pathohistology. Results All rats survived after administering ip for 26 weeks but food-intake of rats with high dosage was decreased, weight enlarged slowly, aspartate aminotransferase(AST) increased and clotting time (CT) prolonged. Some hepatic injuries were seen in high dosage group, but these symptoms were relived or disappeared after withdrawal. No evident toxicity reaction was seen in the medium and low dosage groups. Conclusion By long-term administering ip SGI in rats, the high dosage[ 12.5g/(kg·d)] can restrain weight's increase and hepatic cells in some animals( 7/22 )or generate granular degeneration and denaturation. Liver cells of some rats(4/22) with the medium dosage group appeared low-grand granular degeneration but the hepatic injury could be recovered. The results show that the target organ is liver with long-term administering ip SGI and the hepar of each rat doesn' t find evident injury with withdrawal.
