摘 要:目的探讨细胞间粘附分子(ICAM-1)表达与中性粒细胞(PMNs)浸润、缺血再灌注损伤的关系,并观察水飞蓟宾-卵磷脂复合物(silybin-lecithin compound,SLC)对缺血再灌注损伤的保护作用机理.方法蒙古沙土鼠155只.分设:1)缺血组;2)缺血再灌注组;3)SLC100mg/kg组;4)SLC200mg/kg组;5)SLC400mg/kg组;6)假手术对照组;7)溶剂组.除假手术对照组外,每组分设1、3、6、12、24h时相点.取鼠脑组织块用原位杂交和免疫组化检测方法测定ICAM-1mRNA及其蛋白表达水平,用酶法测定PMNs浸润数.结果脑缺血再灌注时,ICAM-1表达明显增高,其中ICAM-1mRNA表达至6h达高峰,而ICAM-1蛋白质表达水平、PMNs浸润数改变于12~24h达最高峰,二者之间呈显著正相关,但ICAM-1mRNA与后二者间无明显相关性.SLC组上述指标于再灌注时显著下降,其下降程度与时相和浓度呈明显的负相关.均P<0.01或0.05.结论脑缺血再灌注时,ICAM-1参与介导了PMNs对组织细胞的粘附、浸润和再灌注损伤的发生发展,SLC可通过抑制ICAM-1的表达而产生神经细胞的保护作用.Objective To investigate the influence of expression of intercellular adhesion molecule -1 (ICAM-1) on cerebral isehemie reperfusion injury and cerebral infiltration of polymorphonuclear neutrophils (PMNs). Methods using an established common carotid artery occlusion model.we observed the expression of ICAM- 1 by in setup hybridization and the protein by immunocytochemistry, and the numbers of infiltrated PMNs were measured with MPO enzyme marker method. Results After cerebral isehemie reperfusion,eerebral levels of expression of ICAM- 1 were increased significantly and reached a peak at 6h;but the ICAM- 1 protein peaked during 12-24h. Number of PMNs also increased and peaked after reperfusion 12-24h. Close positive correlation (P 〈 0.05) was found between the levels of expression of ICAM- 1 and PMNs. In SLC group,all of the indices were increased after isehemie reperfusion,but the levels of increase were significantly lower (P 〈 0. 05 or P 〈 0.01) as compared with PVP group. Conclusions The findings indicate that ICAM-1 and PMNs is implicated in the development of cerebral isehemie reperfusion injuries. SLC could protect the cerebral tissue from cerebral isehemie reperfusion by partially blocking the expression of ICAM- 1.
