Simultaneous determination of berberine hydrochloride and palmatine hydrochloride in Cortex Phellodendri chi- nensis by capillary electrophoresis

摘  要:目的建立同时测定黄柏药材中盐酸小檗碱和盐酸巴马汀含量的方法。方法采用高效毛细管电泳法。运行缓冲液为50mmol·L-1Na2B4O7-CH3OH(85:15),pH7.0;操作电压14kV;检测波长265nm。结果在选定的电泳条件下,盐酸小檗碱和盐酸巴马汀的线性范围分别为0.005-0.08和0.002-0.03g·L-1,r=0.9997和0.9985;方法的加样回收率分别为98.3%和99.4%,精密度分别为1.89%和2.21%。结论所建立的分析方法具有准确、简便、快速的特点,可以用于黄柏药材中盐酸小檗碱和盐酸巴马汀的同时分析检测。OBJECTIVE To establish an analytical method for simultaneous determination of berberine hydrochloride and palmatine hydrochloride in Cortex Phellodendri chinensis. METHODS High performance capillary electrophoresis was used with the mobile phase consisting of 50 mmol·L^-1 sodium tetraborate solution-methanol (85:15, pH 7.0). The applied voltage was 14 kV and detection wavelength was at 265 nm. RESULTS The linear curves for berberine hydrochloride and palmatine hydrochloride were obtained in the range of 0.005 - 0.08 mg· mL^ -1 ( r = 0.999 7) and 0.002 - 0.03 g· L^ - 1 ( r = 0.998 5 ), respectively. The recovery was 98.3 % with RSD of 1.89 % for berberine hydrochloride and 99.4% with RSD of 2.21% for palmatine hydrochloride. CONCLUSION The method can be applied to the simultaneous analysis of berberine hydrochloride and pahnatine hydrochloride in Cortex Phellodendri chinensis.
