摘 要:目的:研究注射用清开灵冻干粉解热、镇痛作用及其解热机制。方法:建立家免2,4-二硝基苯酚及热模型及大鼠内毒素发热模型,观察清开灵冻干粉解热作用;采用小鼠扭体法及热板法,观察清开灵冻干粉镇痛作用。并用放免法检测大鼠下丘脑cAMP及IL-1β含量的变化。结果:注射用清开灵冻干粉能有效抑制内毒素导致的大鼠发热和2,4-二硝基苯酚导致的家兔发热,显著降低发热大鼠下丘脑IL-1β及cAMP的含量;明显减少醋酸致痛小鼠扭体次数,推迟热致痛小鼠添后足时间。结论:注射用清开灵冻干粉具有解热、镇痛作用,其解热机制可能通过抑制内生致热原IL-1β的生成,从而降低下丘脑发热介质cAMP的含量。Objective: To investigate the antipyretic, analgesic effects of Qingkailing Freeze-Dried Powder for Injection(QI) and its mechanism of the antipyretie effect. Methods: The rabbit model of 2,4-dinitrophenol-induced fever and the rat model of endotoxininduced fever were established and the body temperatures were observed. Radioimmunoassay was adopted to detect the levels of IL-1β and cAMP in the hypothalamus of rats. The analgesic effects were tested by hot-plate and writhing reaction method. Results: QI showed marked antipyretie effects on the 2.4-dinitrophenol-induced fever and the endotoxin-induced fever. At the same time, Q1 remarkably decreased the contents of IL-1β and cAMP in the hypothalamus. Q1 also remarkably reduced the frequency of acetic acid-induced writhing and delaied the time of licking rear feet. Conclusion: Q1 has antipyretic, analgesic effects. Its mechanism of the antipyretie effects on rats may be inhibiting the production of IL-1β and accordingly decreasing cAMP in the hypothalamus.
