桃红四物汤及其拆方对血管生成的影响

摘　　要：目的观察桃红四物汤及其不同药物组合对血管生成的作用。方法种鸡蛋孵化7d建立鸡胚绒毛尿囊膜(CAM)模型,加药于两条主干血管之间的相对无血管区。孵化第10d检测CAM载体周围5mm的血管数目(N)、血管面积(A)和血管面积相对比值(AA)。结果桃红四物汤Ⅰ号、Ⅱ号、四物汤Ⅱ号、桃仁红花、桃仁、当归、川芎、赤芍组CAM的细小血管N、A及AA显著减少,而四物汤Ⅰ号、熟地白芍、熟地组CAM的细小血管N、A及AA均显著增加。结论桃红四物汤及其拆方中活血组别,即桃红四物汤Ⅰ号、Ⅱ号、四物汤Ⅱ号、桃仁红花、桃仁、当归、川芎、赤芍组具有抗CAM血管生成作用;而补血组别,即四物汤Ⅰ号、熟地白芍、熟地组具有促CAM血管生成作用。Objective： To study the effects of Taohongsiwu Decoction and its separated prescriptions decoction on the angiogenesis of chick chorioallantoie membrane（CAM） in vivo. Methods： 7day-old fertilized white chicken egg incubated at 37℃ in charnbe was prepared by creating a window in the shell. A sponge carrier was placed on CAM. The experimental groups were treated with Taohongsiwu Ⅰ, Ⅱ, Siwu Ⅰ , Ⅱ, Taorenhonghua, Shudibaishao （（桃红四物汤Ⅰ、Ⅱ,四物Ⅰ、Ⅱ,桃仁红花,熟地白芍） and eight single herb decoction at a concentration of 2g/ml （20μl per egg, dayly, 3 days）. On day 10, CAMs around the carriers 5mm were investigated. The amount of blood vessel（N）were counted using a stereomicroscope. The blood vessel＇s area（A）and the ratio between the vessel area and the CAM area（AA）were analyzed by computer image analysis systerm. Results： Taohongsiwu Ⅰ, Ⅱ, SiwuⅡ, Taorenhonghua, Taoren, Danggui, Chuanxiong and Chishao decoction （（桃红四物Ⅰ,Ⅱ,四物Ⅱ,桃仁红花,桃仁,当归,川芎和赤芍） groups were obviously inhibited the N, A and AA of the CAM, on the contrary, Siwu Ⅰ, Shudibaishao and Shudi （四物Ⅰ,熟地白芍和熟地） decoction groups can obviously stimulated the N, A and .Gat of the CAM（ P〈 0.05）. Conclusion： The promoting blood flow prescriptions and herbs such as Taohongsiwu Ⅰ, Ⅱ, Siwu Ⅱ, Taorenhonghua, Taoren, Danggui, Chuanxiong and Chishao decoction groups have potent activity to inhibit the CAM angiogenisis, whereas the invigorating blood prescriptions and herbs such as Siwu Ⅰ, Shudibaishao and Shudi decoction groups can obviously stimulate the CAM angiogenisis.
