摘  要:对采自中国东海外海和日本海的黄鲷Taius tumifrons线粒体细胞色素氧化酶Ⅰ亚基(COⅠ)基因进行了研究。利用通用引物对该序列进行了PCR扩增,去掉两端引物及部分序列共得到590bp的碱基片断,其中A、T、G、C平均含量分别为30.4%、24.3%、25.0%和20.3%,A+T(54.7%)高于G+C(45.3%)含量。在获得的9个序列中共发现有156个碱基存在变异,其中包括42处简约信息位点,没有发现碱基的插入和缺失,序列中的转换大于颠换。实验结果显示,个体间的遗传距离在0—0.223 6之间,c3和j3与其它个体相比遗传差异明显。2群体间平均遗传距离为0.089 7,而东海和日本海群体内的平均遗传距离分别为0.086 9和0.113 3,说明黄鲷个体间遗传差异较大,但群体间的遗传差异不明显。The 590bp fragments of cytochrome oxidase subunit Ⅰ (CO Ⅰ ) gene were obtained by PCR method from mitochondrial DNA of Talus tumifrons from two local populations (the East China Sea and the Sea of Japan) using universal primers. The A, T, G, C average contents of sequences were 30.4%, 24.3%, 25.0% and 20.3%, respectively, and the content of A+T (54.7%) was higher than that of G+C (45.3%). The sequences showed 156 substituted sites (26. 4%), including 42 parsimony-informative sites, and the transition times were longer than transversion times. No insertion and deletion of sites were found. Genetic distances between the individuals varied from 0 to 0. 223 6. High levels of genetic variation were found in two individuals (c3 and j3). The average genetic distances were 0. 089 7 between the two local populations, and 0. 086 9 and 0. 113 3 within the populations of the East China Sea and the Sea of Japan, respectively, revealing that genetic variance was obvious between individuals but unobvious between the two local populations.
