摘 要:目的:研究黄芪对102速殖子感染小鼠肝脏的影响,探讨黄芪对急性弓形虫感染小鼠的保护作用机制。方法:建立102速殖子感染小鼠模型,设黄芪治疗组和对照组。2组小鼠定期分批处死,取血清和肝脏。用荧光定量PCR检测感染后第4、8天小鼠肝脏的虫荷。用赖氏法测定感染后第4、5、6、7天血清丙氨酸转氨酶(ALT)水平。取感染后第2、4、6、8天肝脏做冷冻切片,黄递酶组织化学染色方法进行iNOS酶组化染色。结果:感染后第4天和第8天肝脏虫荷两组差异无显著性。感染后黄芪组和对照组血清ALT水平持续上升,黄芪组感染后第4天为87.20U(卡门单位),显著高于对照组(34.44U,P<0.05);感染后第8天为172.50U,显著低于对照组(225.69U,P<0.05)。对照组iNOS表达局限于小叶中央,汇管区浅染。结论:黄芪对急性弓形虫感染小鼠的肝脏功能存在双向调节的作用,可能与其诱导iNOS的作用有关。Objective:To evaluate the effects of Astragalus membrane on mice lever infected with 100 tachyzoites of virulent RH strain of Toxoplasma Gondii, and to study the mechanism of the protection of Astragalus membrance on mice of acute toxoplasmoslis. Methods: Mice infected with 10^2 tachyzoites were orally treated with Astragli membrane and the control groups were treated with water. Mice were killed to collect the blood sera and livers respectively on a regular basis. Parasites loads at 4 and 8 dpi (day post infection) were determined with quantitative fluorescence Poly merase Chain Reaction (PCR).Serum enzyme levels of alanine aminortransferase (ALT) at 4,5,6,7 dpi were assayed. Frozen sections of lever at 2, 4, 6, and 8 dpi were stained by the method of nicotinamide adenosine dinudeotide phosphate diaphorase to quantitate iNOS. Results: No significant difference of parasite loads in livers and lungs at 4 dpi and 8 dpi were demonstrated between Astrgli group and control group. The ALT levels in sera of both groups kept elevating. ALT level was observed increasing to 87.20 U at 4 dpi in Astrgali group, while it remained low (34.44 U)in control group. But at 8 dpi, ALT level in Astrgali group (172.50 U)were significant lower than that in control group (225.69 U)(P 〈 0.05). Control group demonstrated low levels of iNOS expression until death, while Astragli induced higher levels of iNOS from 4 dpi and more extensive expression at both portal tracts and aroud the central vein. Conclusion: Astragalus membrance can exert its deliberate regulation on the livers of mice of acute toxoplasmosis, possibly due to its function of inducing iNOS.
