柑成熟种子胚培养获得四倍体植株

摘　　要：通过组培方法从374粒椪柑种子培养中获得449株小苗。利用流式细胞仪进行染色体倍性分析,发现其中7株小苗为四倍体植株。采用13对SSR引物分析表明这7株四倍体植株均来自珠心细胞,为同源四倍体。这些四倍体的获得为下一步培育三倍体无核品种奠定了基础。Seedless or few seed trait is one of the goals of Ponkan breeding. By culturing the embryos of mature seeds, a total of 449 polyembryonic seedlings from 374 seeds were obtained; and seven of them were verified to be tetraploid by flow cytometry. SSR （simple sequence repeat） analyses with 13 primers indicated that these tetraploid seedlings were all autotetraploid from the nucellar cells. These tetraploid seedlings will be of great value for creating seedless Ponkan through sexual hybridization.
