摘 要:目的通过巨噬细胞、雪旺细胞及感觉神经元的联合培养,了解雪旺细胞的活性及感觉神经元的存活。方法(1)取乳鼠坐骨神经进行雪旺细胞的分散培养,24h后在腹腔中取巨噬细胞并用分隔培养皿进行联合培养。(2)联合培养后24h、48h、72h,采用RT-PCR法,半定量分析雪旺细胞NGFmRNA含量的变化。(3)将巨噬细胞和雪旺细胞的联合培养基加入至乳鼠背根神经节感觉神经感觉神经元体外培养中,培养48h后观察神经元的生长情况,用MTT法测定神经元的活性。结果单纯雪旺细胞培养中存在NGFmRNA但表达量较低,加入巨噬细胞后NGFmRNA表达量显著升高。联合培养条件培养基可明显促进体外培养的体外培养中感觉神经感觉神经元的存活。结论巨噬细胞可改变雪旺细胞的活性,联合培养条件培养基对感觉神经感觉神经元有明显的营养作用。Objective To investigate the activity of Schwann cells and the survival of dorsal root ganglion sensory neurons by co-culture of macrophages, Schwann cells and sensory neurons, Methods Schwarm cells from the sciatic nerve of natal rats were cultured for 24 hours and then they were co-cultured with macrophages obtained from the rat's abdominal cavity in a divided culture dish. After 24, 48 and 72 hours, the concentration of NGF mRNA in Schwann ceils was analyzed by RT-PCR. The culture medium collected from the dish containing the macrophages and Schwann cells was added to the sensory neurons in vitro. The growth and survival of sensory neurons were observed and measured by MTT method. Results Pure Schwarm cell culture may produce NGF mRNA but its concentration was obviously raised after macrophages were added. Co-culture conditioned medium may markedly promote survival of the sensory neurons in vitro. Conclusion Macrophage may change the activity of Schwann cells and co-culture conditioned medium can provide good nutrition to the sensory neurons.
