摘 要:目的:研究针对c-myc mRNA不同位点的两种反义寡脱氧核苷酸序列(AS-ODN):myc-AUG AS-ODN,myc-CRD AS-ODN,对类风湿滑膜细胞增殖的抑制作用。方法:培养人类风湿滑膜B型细胞,合成反义寡脱氧核苷酸序列并进行硫代磷酸化修饰,以脂质体L ipofectin为载体,转染滑膜细胞,用MTT法检测细胞的增殖情况。结果:myc-AUG AS-ODN,myc-CRD AS-ODN均能抑制滑膜细胞增殖,使细胞数目减少的最大值分别为40.0%和41.4%。myc-AUG AS-ODN抑制滑膜细胞增殖的起效时间早于myc-CRD AS-ODN。在高浓度时寡脱氧核苷酸具有非序列特异性细胞毒作用。结论:两种AS-ODN序列均可用于抑制滑膜细胞增殖。Objective: To investigate the difference in inhibiting synoviocytes proliferation by 2 c-mye antisense oligodeoxynucleotides (AS-ODN) sequences, myc-AUG AS-ODN and myc-CRD AS-ODN. Methods: Human synoviocytes from patients with rheumatoid arthritis were cultured. The sequences were modified by phosphorothioates. Lipofectin was used as a acarrier. MTT assay was used to examine the inhibition of cell proliferation. Results: Both myc-AUG AS-ODN and myc-CRD AS-ODN inhibited synoviocytes proliferation. The maximal decrement of cell number were 40. 0% and 41.4%, respectively. The inhibiting time of myc-AUG AS-ODN was shorter than that of myc-CRD AS-ODN. ODN of high level had non-sequence-specific cytotoxicity. Conclusion: Both sequences of c-myc AS-ODN are useful in inhibiting synoviocytes proliferation.
