摘 要:目的:考察二仙汤在煎煮过程中淫羊藿苷含量的变化规律.方法:采用HPLC检测方法,色谱柱为Hypersil-RDSC18(4.6 mm×200 mm,5μm);流动相为甲醇:水:冰乙酸(80:70:2),流速为1 mL/min;检测波长为270 nm.同单味药材比较,观察不同煎煮时间对二仙汤中活性成分淫羊霍苷含量的动态变化规律.结果:淫羊藿苷的线性范围为0.51~51μg/mL,r=0.999 9.低、中、高浓度的加样回收率分别为(101.36±2.60)%、(99.17±1.17)%、(100.82±1.89)%(n=5).二仙汤中淫羊藿苷含量随煎煮时间延长而逐渐减少,最终达到动态平衡.结论:煎煮时间直接影响煎煮液中淫羊藿苷的含量,制备过程应避免过热.Objective: To study dynamic content changes of icariin in Er-xian decoctum in the decoction process. Methods: The content of icariin was determined by HPLC method. Hypersil-RDS C18 (4.6 mm×200 mm, 5 μm) column was used, with methanol : water : acetic acid(80 : 70 : 2) as the mobile phase and 1 mL/min as the flow rate. The detection wavelength was 270 nm. Transformation principle of icariin in Er-xian decoctum was investigated in different boiled time compared with single crude drug. Results: The linear range of icariin was 0.51-51 μg/mL, r=0.9999, and the recoveries in low, middle and high concentrations were (101.36±2.60)% ,(99.17±1.17) % ,(100.82±1.89) % (n=5), respectively. Content of icariin in Er- xian decoctum was decreased as boilded time increased, and finally reached a kinetic balance. Conclusion: The time of decoction can directly influence the content of icariin. Over-heating should be avoided in the whole process.
