摘  要:目的:探讨流式细胞术中细胞周期常规分析方法的误区和潜在的合理分析方法。方法:设立空白对照,观察MOLT-4细胞经0.1μmol·L-1喜树碱诱导细胞周期阻滞7h后细胞周期变化情况,并从常规DNA直方图分析法(方法一)和本实验意外发现的方法(方法二)进行细胞周期分析,并评价两种方法获得的结果。结果:方法一分析的细胞周期分布结果G0/G1期、S期和G2/M期分别为(65.42±4.73)%、(25.01±2.58)%、(9.57±1.96)%,方法二分析结果分别为(31.51±2.13)%、(59.32±5.92)%、(9.17±1.61)%。两种分析方法获得的结果存在显著差异(n=8,P<0.01)。结论:流式细胞术常规细胞周期分析方法存在严重的误区。一种新的基于细胞周期特异性标志物的定量技术或形态学方法应用于细胞周期的分析较合理。Objective: To evaluate a mistake of routine method of cell cycle analysis in flowcytometry (FCM) and a possible better method in analyzing cell cycle distribution. Methods: MOLT-4 ceils were exerted with induction of cell cycle arrest by 0. 1μmol · L^-1 camptothecin for 7 hours. Changes of cell cycle distribution were observed. Method 1 of routine DNA histogram analysis and Method 2 of unexpected discovery in this experiment were used to analyze cell cycle distribution. Then results from two methods would be assessed. Results: The percentages of G0/G1 phase, S phase and G2/M phase analyzed by Method 1 were (65.42±4.73)%, (5.01±2.58)% and (9. 57±1.96)%, respectively. However, with Method 2, the results were respectively (31.51±2.13)%, (59.32±5.92)% and (9. 17±1.61)%. Results obtained from Method 1 and Method 2 were significantly different(n=8,P〈0.01). Conclusion: There is a mistake in routine cell cycle anlysis in FCM. A novel method of quantificational technology or morphological evidence based on cell cycle-specific indicators is more of rationality in cell cycle analysis.
