摘　　要：目的探讨辐射损伤对巨核细胞调控因子IL-6和TNF-α表达的影响。方法用RT-PCR和W estern b lot方法检测辐射后Dam i细胞IL-6、TNF-α表达水平的变化。结果辐射损伤3、5、10 Gy后,IL-6的mRNA水平和蛋白水平表达均显著低于对照组(P<0.05),其变化趋势是随照射剂量增加而表达逐渐降低。TNF-α在mRNA水平变化无显著性差异,但在蛋白水平表达则显示,3个剂量组均显著高于对照组(P<0.05),并随剂量增加表达逐渐升高。结论不同剂量放射损伤所致造血功能障碍与巨核细胞调控因子IL-6和TNF-α的异常表达有关。Objective To analyse the expression of IL-6, TNF-α in Dami cells after exposure to ^60Co γ- radiation. Methods Dami cells irradiated with ^60Co γ-rays were usded to analyze the IL-6 and the TNF-α expression by RT-PCR and Western blotting in 24 h after irradiation. Normal Dami cells were used as control. Results The expression of IL-6 in Dami cells after irradiation at the dose of 3 Gy, 5 Gy, and 10 Gy was lower than that in normal Dami cells （ P 〈 0. 05 ）. The protein expression of TNF-α in Dami cells after irradiation was higher than that in normal Dami cells （P 〈 0. 05）. The expression of IL-6 and TNF-α showed a direct correlation with the dose. Conclusion γ-radiation inhibits IL-6 expression but promotes TNF-α expression, suggesting that the abnormal expression of IL-6 and TNF-α in Dami cells is involved in the injury of hemopoiesis by γ- radiation.
