摘 要:研究高斯光源的束斑半径、光源孔径和探测器孔径对荧光共焦显微镜3-D OTF的影响,获得了具有有限光源孔径、探测器孔径和束斑半径的荧光共焦显微镜的三维光学传递函数(3-D OTF)。数值计算结果表明,束斑半径影响到光源孔径和探测器孔径的选择,与采用平行光比较,采用小束斑半径的高斯光束可以提高光源孔径和探测器孔径,获得更好的分辨率和更高的信噪比。The influence of the three parameters,namely the beam size of Gaussian source, the source size and the detector size,on the three-dimensional optical transfer function (3-D OTF) of an incident light fluorescent confocal microscope is investigated, and the 3-D OTF of such a set-up is derived. The calculated results show that the beam size has an effect on choosing both the source size and the detector size by means of a gaussian beam with the smaller beam size, both the source size and the detector size can be increased, and better optical resolution and higher signal-noise-ratio can be reached, compared with those by means of a parallel beam.
