摘 要:目的研究电针治疗对成年大鼠脑缺血后缺血侧神经干细胞巢蛋白(nestin)表达的影响,探讨电针治疗缺血性脑损伤的作用机制。方法采用开颅热凝闭法制作大脑中动脉阻塞(MCAO)模型,随机分为模型组与电针组,针刺“大椎”、“百会”,并行电针干预,采用巢蛋白免疫组化法观察脑缺血后3,7,14与21d四个不同时相巢蛋白表达阳性细胞的数量。结果脑缺血后不同时相缺血侧皮质、海马齿状回和纹状体均有巢蛋白表达阳性细胞,其中7d时的阳性细胞数量明显增加(皮质:P<0.05,海马:P<0.01,纹状体:P<0.01);而电针组在不同时相的巢蛋白阳性细胞数量均较同时相模型组有明显的增加(P<0.05或P<0.01)。结论电针可促进缺血后相关脑区巢蛋白阳性细胞数量的增加,提示这种作用可能是电针治疗脑缺血的重要作用机制之一。Objective To study the influence of electroacupuncture on the expression of Nestin in neural stem cells after focal cerebral ischemia in rats and to explore the mechanism of electroacupuncture in treating ischemic brain injury. Methods The model of middle cerebral artery occlusion was made by using opening cranium hot coagulation in 72 rats, who were divided randomly into a model group and an electroacupuncture group. Electroacu- puncture was applied on “ Dazhui” and “ Baihui” points in rats in electroacupuncture group, and immunohistochemistry method was used to observe the number of Nestin expression positive cells on the 3th, 7tb, 14th and 21st days after ischemia. Results There were Nestin positive cells in the cortex, hippocampal dentate gyrus (DG) and striatum at different time points after cerebral ischemia, and the number of Nestin positive cells in various brain regions were increased significantly on the 7th day (P 〈 0.05). It was also found that the number of Nestin positive cells in electroacupuncture group was significantly more than that in the model group at different time points ( P 〈 0.05 or P 〈 0.01 ). Conclusion Electroacupuncture can increase the number of Nestin positive cells in some brain regions after focal cerebral ischemia, which might be one of the important mechanisms of electroacupuncture in treating ischemic brain injury.
