摘 要:目的观察大鼠背根神经节(DRG)中即刻早期基因(IEG s)c-jun在慢性压迫性损伤(CC I)疼痛模型中的变化,探讨慢性神经痛的发生机制。方法将SD大鼠18只,随机分成CC I组和假手术组各9只,CC I组参照B ennett和X ie等方法建立CC I模型,假手术组仅暴露坐骨神经主干,不结扎。于术前2d和术后1、3、5、7d测定两组机械痛阈和热痛阈,术后7d取大鼠双侧L4、L5和L6DRG,免疫组化法分析DRG c-jun变化。结果CC I组损伤侧于术后3d出现热痛觉过敏(P<0.05),术后7d达高峰(P<0.01),且DRG c-jun出现明显表达;而健侧和假手术组没有表达(P<0.01)。结论初级感觉神经元中c-jun表达是神经损伤的标志和始动因素。Objective: To investigate the changes of c-jun in dorsal root ganglion (DRG) neurons in chronic constriction injury (CCI) model of chronic neuropathic pain. Methods: Eighteen rats were divided into CCI group and control group with 9 animals in each. The CCI model was made by loose ligation of sciatic nerve with chronic gut. The thermal pain threshold was measured before operation and 1,3,5,7 days after operation. The animals were deeply anesthetized and rapidly decapitated 7 days after surgery. The L4-L6 DRG of the operated side were removed. The changes of c-jun expression was determined by immunochemistry. Results: Pain threshold in CCI group was significantly lower than that in control group. The immediate early gene (IEGs) c-jun in creased to the peak 7 days after CCI as compared with that in control group (no expression was detected). Conclusion: The IEGs c-jun is considered as the marker of nerve injury and involved in the hyperexcitability of the primary sensory neurons after CCI.
