摘  要:目的:观察逆针关元穴对自然更年期雌性大鼠抗衰老指标的调节作用。方法:实验于2004-04在北京中医药大学针灸学院针灸机理实验室完成。选取健康9.5月龄SD雌性大鼠56只为实验动物。选择与人类更年期综合征病理变化更为近似的自然老化术制备模型。采用随机抽签的方法将实验动物分成2组,自然对照组和逆针组。自然对照组按照自然月龄分为4组,自然10,12,14,16月龄组,每组8只。各组每天只进行抓取刺激,不作针刺。逆针组在与自然组一同养至10月龄后,采用逆针大鼠关元穴,在关元穴下约2分处将针直刺2分,后斜向上完全刺入,留针20min。留针过程中让大鼠自由活动。2次/周,持续针刺8周(2个月),并于进入更年期的12,14,16月龄后分别取材,分别命名为逆针12,14,16月龄组。每年龄组8只。自然对照组与逆针组同批取材。血液标本均于上午8:00~11:00采集。经处理后采用免疫比浊法测定血浆和子宫超氧化物歧化酶、一氧化氮合酶水平,按操作说明书进行。各组组间差异采用单因素方差分析。结果:56只实验大鼠均获得检测,无缺失值。①血浆超氧化物歧化酶水平:自然组14,16月龄比自然组10月龄显著降低犤(133.72±29.16),(149.04±27.74),(221.78±15.48)nkat/L,P<0.05犦。逆针组14月龄与同龄自然组比较明显升高犤(249.75±45.50),(133.72±29.16)nkat/L,P<0.05犦。②血浆一氧化氮合酶水平:自然组14月龄与自然组10月龄比较,呈显著降低趋势犤(12.21±0.84),(18.07±1.05)nkat/L,P<001犦。逆针组14月龄与自然组14月龄比较明显升高犤(21.90±1.71),(12.21±0.84)nkat/L,P<001犦。③子宫超氧化物歧化酶水平:自然组12,14,16月龄比自然组10月龄显著降低(P<0.05)。逆针组12,16月龄比自然同月龄组显著上升(P<0.05或0.01)。④子宫一氧化氮合酶水平:自然组14,16月龄与自然组10月龄比较,呈显著降低趋势(P<001)。逆针组12、16月龄与同年龄自�AIM:To research the regulation acupuncting Guanyuan point on the climacteric rats. mechanism of the effects of preindex of antidecrepitude in the natural METHODS: This experiment was conducted in the Mechanism Laboratory of Acupuncture and Moxibastion , Beijing University of Traditional Chinese Medicine in April 2004. Totally 56 female healthy SD rats aged 9.5 months were recruited. Naturally aging models similar to human pathological change of climacteric syndrome were chosen. The animals were randomly divided into 2 groups with simple random sampling method, namely normal control group and pre-acupuncture group. The rats in the normal control group were divided into 4 groups according to natural months, namely 10-month group, 12-month group, 14-month group and 16- month group with 8 rats in each group. Animals in each group were performed grasping stimulation without needle acupuncture. The animals in the pre-acupuncture group were raised together with the animals in the control group for 10 months, performing acupuncture 2 scores at 2 scores of Guanyuan point, then oblique complete acupuncture, retaining the needle for 20 minutes. Rats can move freely during retaining the needle. Twice per week for consecutive acupuncture for 8 weeks (2 months) and materials were collected 12, 14 and 16 months after entering menopause and namely 12-month group, 14-month group and 16-month group respectively with 8 rats in each group. The materials in the normal control group and pre-acupuncture group were collected at the same time. And blood sample was collected at 8:00-11:00 AM. Immune turbidimetry method was used to measure the superoxide dismutase (SOD)and nitricoxide synthase (NOS)in the blood and uterus according the instruction. Single-factor analysis of varience was used for comparing the difference among the groups.
