摘 要:根据传染性喉气管炎病毒(ILTV)美国632株TK基因序列设计并合成1对引物,以ILTV烟台株DNA为模板扩增了TK基因,并对其进行了序列测定.将ILTV烟台株的TK基因和TK蛋白分别与ILTV美国632株,英国Thorne株,Beijing E2株,BHV-1,EHV-1,EHV-2,FHV-1,HHV-1,HHV-2,HHV-3,HHV-4,HVT,MDV-1,MDV-2,PRV和SHV-2的TK基因和TK蛋白比较后发现,其TK基因的核苷酸和TK蛋白的氨基酸同源性分别为24.6%~99.5%和15.1%~98.9%,表明不同ILTV毒株之间TK基因和TK蛋白相对保守,但与其他α-疱疹病毒的TK基因和TK蛋白同源性则较低.A pair of primers flanking the TK gene were designed according to the published nucleotide sequence of the ILTV USA 632 strain. The thymidine kinase (TK) gene of the wild Chinese ILTV Yantai strain was obtained by polymerase chain reaction and the PCR products were cloned into pMD18-T vector and the nucleotides of the TK gene was sequenced. By analyzing with DNA Star software,we found that the homology of the TK gene and the deduced amino acid of ILTV Yantai strain with that of ILTV USA 632 strain, England Thorne strain, Beijing E2 strain, BHV-1, EHV-1, EHV-2, FHV-1, HHV-1, HHV-2, HHV-3,HHV-4,HVT,MDV-1,MDV-2,PRV and SHV-2 were between 24.6%-99.5% and 15. 1%- 98.9%. The results showed that the TK gene was very conserved among different ILTV strains,but very low between different herpesvirus.
