摘 要:目的:观察大黄治疗组和正常组小鼠肠道免疫分泌物的变化;探讨大黄对肠道黏膜屏障保护作用的机制。方法:实验用BALB/c小鼠。分成生理盐水灌胃组和10%大黄煎剂灌胃组。24h后处死动物,灌洗肠道,收集灌洗液,分别测定IgA、总蛋白、补体C3、高密度脂蛋白(HDL)、Ⅱ型磷脂酶A2(PLA2)活力和溶菌酶的含量;同时切取并收集各小鼠小肠组织40mg,运用RT-PCR扩增,琼脂糖凝胶电泳成像检测隐窝素-1基因和隐窝素-4基因扩增片段含量。结果:2组小肠灌洗液中IgA,总蛋白,补体C3,溶菌酶含量以及Ⅱ型PLA2活力有统计学差别(P<0.05)。2组小肠灌洗液中HDL含量无统计学差别(P>0.05)。2组小肠组织匀浆中隐窝素-1(Cr1)基因扩增片段含量与隐窝素-4(Cr4)基因扩增片段含量无统计学差别(P>0.05)。结论:大黄能促进肠黏膜上皮分泌多种免疫相关物质,对于减轻创伤、烧伤、休克等严重应激反应时的肠黏膜损伤,防止肠道菌群移位和全身炎症反应综合征的发生具有重要的理论和临床意义。Objective: To investigate the therapeutic mechanism of rhubarb in protecting the intestinal muco-membranous barrier in the mice. Method. Bal b/c mice were divided into 2 groups, gavaged with normal saline and 10% rhubarb decoction, respectively. The animals were killed after 24 hours after the treatments. The intestinal juice was collected after intestinal lavage and centrifuged for determination of IgA, total protein, C3, high density lipoprotein, type Ⅱ PLA2 activity, and content of lysozyme. At the same time, 40 mg of small intestine were incised in each mouse.Reverse transcription polymerase chain reaction (RT-PCR) and gel image analysis were performed to detect the content of the cryptdin gene expression. Result: The content of IgA, total protein, the C3, lysozyme, and the type Ⅱ PIA2 activity in intestinal lavaged juice exhibited the statistical differences between the two groups( P 〈 0.05). There were no significant difference in the ontents of HDL, cryptdin-1 and cryptdin-4 gene expression between the two groups ( P 〉 0.05). Conclusion: Rhubarb could increase secretion of several immune associated substances of the mucous membrane in normal intestine, indicating a possibility to abate the injury of intestine mucus resulted from severe stress induced by trauma,bum and shock. Through above mechanisms Rhubarb may also reduce the incidence of bacterial translocation and systemic inflammatory reaction syndrome (SIRS),
