摘 要:目的研究与HIV-1感染相关的趋化因子受-体2(chemokine receptor-2,CCR2)等位基因变异频率和多态性在中国达斡尔族、布依族人群的分布特征。方法应用PCR-RFLP等方法检测两个人群共132个个体中CCR2-64I基因变异频率。结果达斡尔族、布依族人群中CCR2-64I基因频率分别为0.2647和0.1172,且存在着较大的差异(χ2=9.207,P=0.002)。两个人群CCR2-64I等位基因群体分布符合Hardy-Weinberg平衡。结论获得了达斡尔族、布依族人群CCR2-64I基因频率,为评估中国人群对HIV-1的遗传易感性及艾滋病的流行病学研究提供了基本数据。Objective To investigate the frequencies and polymorphisms of HIV-1 resistant CCR2-64I alleles in Chinese Daur and Bouyei populations. Methods Whole blood samples from 68 Daur subjects and 64 Bouyei subjects were collected randomly and their genomic DNA were extracted. Allelic frequencies were identified by PCR-RFLP analysis. Results Allelic polymorphisms in the two populations in the samples were analyzed by 2 test. The frequencies of CCR2-64I allele in Daur population and Bouyei population were 0.2647 and 0.1172, respectively. Distributions of the CCR2-64I allele of the two populations were in accordance with Hardy-Weinberg equilibrium, and the frequencies were found significant variance between the two Chinese populations(χ^2=9.207, P=0.002). Conclusion The genotyping and polymorphism of CCR2-64I alleles in Chinese Daur and Bouyei populations are studied for the first time in China, our studies provide referent information for further study of the susceptibility to HIV-1 and the AIDS epidemiology of the Chinese populations.
