摘 要:腺苷一磷酸脱氨酶(adenosine monophosphate deam inase,AMPD)是嘌呤代谢过程中的一种重要的酶,它对肉质性状和风味起到重要作用。根据人和大鼠的AMPD1基因序列设计引物对鸡的AMPD1基因进行扩增,在克隆测序的基础上,采用PCR-RF-SSCP技术分析了AMPD1基因在北京油鸡(119只)、泰和乌骨鸡(40只)、崇仁麻鸡(39只)、鹿苑鸡(39只)、霞烟鸡(40只)和AA鸡(40只)6个鸡种中的多态性。结果表明,在供试的6个鸡种中共检测到AMPD1基因的A、B、C 3个等位基因,产生AA、AB、AC、BB、BC、CC 6种基因型。测序结果发现等位基因B和等位基因A相比存在一个单碱基突变,即C→T(402),而等位基因C与等位基因A相比存在G→T(361)、A缺失(373)、T缺失(404)、G→T(420)、C→T(434)、G→A(456)、A→G(469)、G→A(473)、G→A(486)、G→A(491)、G→A(493)等11处碱基突变。Adenosine monophosphate deaminase is an important enzyme in purine metabolism, which is essential for meat character and flavor. According to the sequence of adenosine monophosphate deaminase 1 (AMPD1) gene of human and rat, primers were designed to amplify the AMPD1 gene of chicken. Based on the clone sequencing, single nucleotide polymorphism of AMPD1 gene was detected in Beijing fatty chicken(119), Taihe silky chicken (40), Chongren Ma chicken (39), Luyuan chicken (39), Xiayan chicken (40) and Arbor Acres chicken (40) by PCR-RF-SSCP. The results showed that three alleles (A, B,C) and.six genotypes (AA, AB, AC, BB, BC, CC) were found in six chicken breeds. The sequencing results indicated that allele B had only one base difference with allele A (C→T(402)), while allele C had eleven mutations: G→T(361), deletion of A(373), deletion of T(404), G→T(420), C→T(434), G→A(456), A→G(469), G→A(473), G→A(486), G→A(491), G→A(493).
