摘 要:目的研究南江黄羊遗传多态性和探讨AFLP标记在山羊遗传多态性方面应用的可行性。方法应用RAPD和AFLP两种方法对15只南江黄羊个体基因组DNA进行了分析,并比较两种方法的检测效果。结果RAPD标记检测得到的群体遗传相似系数和多态位点比率分别为0.942和26.98%;AFLP标记检测得到的群体遗传相似系数和多态位点比率分别为0.874和31.4%。结论AFLP方法和RAPD均适合于品系内个体间的遗传检测,并且为准确评价南江黄羊的遗传稳定性提供了相关的参数。Objective To study the genetic polymorphism of Nanjiang yellow goat and to investigate the feasibility of this research by AFLP. Methods The genomic DNAs of 15 goats were analyzed with RAPD and AFLP molecule marking techniques. The effects of the two methods were compared. Results The rates of the polymorphic loci and the genetic similar index of population by RAPD marker were 26.98% and 0. 942 respectively, and that by 31.4% AFLP and 0. 874 respectively. Conclusion AFLP and RAPD technique are effective methods in genetic polymorphism monitoring of Nanjiang yellow goats, and supply useful parameter with appraise genetic stability.
