摘  要:目的观察人参总皂甙对放、化疗因素所致骨髓抑制小鼠红系造血所依赖的促红细胞生成素及相应受体的影响。方法采用Co60照射、注射环磷酰胺、氯霉素复合造模,建立骨髓抑制模型小鼠,随机分为给药组和模型对照组,另设正常对照。检测外周血和骨髓有核细胞,并应用造血祖细胞体外培养技术和RT-PCR技术,检测血清Epo和脾脏的EpoRmRNA。结果与正常对照相比,模型小鼠的外周血、骨髓有核细胞和脾EpoRmRNA明显下降,而血清Epo升高(P<0.01)。TSPG组骨髓抑制小鼠的外周血、BMC、血清Epo水平(P<0.01)及脾EpoRmRNA的表达显著高于模型对照组(P<0.05)。结论人参总皂甙可以通过促进机体分泌Epo,并在转录水平调节造血组织相应受体的表达,促进红系造血。Objective To investigate effects of total saponin of panax ginseng(TSPG) on Epo serum concentration and expression of EpoRmRNA in hematopoietic tissue of model mice with myelosuppression. Method Model mice resulted from myelosuppression by irradiation and cytotoxic chemotherapeutic compounds were randomly divided into two groups: TSPG treatment group and model control group. There was an another group as normal control. The count of blood cell and number of BMC were determined by blood auto-analyzer. Serum concentration of Epo were determined by using technique of hematopoietic progenitor cells culture in vitro. Expression of EpoRmRNA in spleen were determined by RT-PCR. Result Compared with normal control group, the count of blood cell , the number of BMC as well as Expression of EpoRmRNA of spleen in model mice decreased and serum concentration of Epo increased evidently(P 〈0.01). All of above indexes in TSPG treatment group were significantly higher than that of model control group(P 〈0.05 or P 〈 0.01 ). Conclusion TSPG can promote erythroid hematopoiesis by enhancing production of Epo and up-regulating expression of EpoRmRNA in hematopoietic tissue of model mice with myelosuppression .
