摘 要:目的研究腹泻患儿的轮状病毒( RV)流行株非结构蛋白 4( NSP4)的基因变异与临床的关系.方 法对昆明地区 2002年和 2003年流行期不同程度腹泻患儿中分离出的 22株轮状病毒流行株的 NSP4基因进行 RT-PCR扩增,并进行 cDNA序列分析,与来自 GenBank Database的 4株人 RV( Wa、 KUN、 AU-1、 Hochi)和 3株动物 RV( EW、 OSU、 SAll)以及我国不同地区 RV流行株 NSP4的基因变异情况进行比较.结果氨基酸同源性表明昆明地区轮状病毒流行株间的同源性高达 98.9%~ 99.4%, 22株流行株全都属于 Wa组.氨基酸进化树提示在 Wa组内又包括 3个亚组.NSP4基因变异与轮状病毒腹泻临床症状严重程度不相关( P>0.05).结论 2002年和 2003年昆明地区轮状病毒流行株皆属 Wa组,且其氨基酸序列极为保守; NSP4基因变异与轮状病毒腹泻临床症状严重程度不相关,但与流行发病的关系需继续研究.Objective To explore the clinical characteristics and variation of the non-structural protein coded by NSP4 genes in infantile diarrhea associated with the outbreak strains of rotavirus infection. Methods NSP4 genes from 22 epidemic strains of human rotavirus isolated in Kunming in 2002 and 2003 were amplified with RT-PCR technique, the resulted cDNA were sequenced. The sequence was analyzed and compared with NSP4 of 4 human rotavirus strains(Wa, KUN, AU-1, Hochi) and 3 animal rotavirus strains (EW, OSU, SAll)from the GenBank Database and the epidemic strains from human rotavirus isolated in different area of China using the Clustal-mp and DNA Ssist and MEGA2 software. Results The homology of the amino acid among the 22 rotavirus strains isolated in Kunming was 98.9% -99.4% ,and all of them were Wa. The Wa group could further be separated into three groups. Variation of genes was found in G3 and G4 serotypes of NSP4 gene, but not in G1. The heavy diarrhea symptom didn' t seem to be associated with NSP4 genetic variations(P〉 0.05) . Conclusions The NSP4 genes from 22 epidemic strains of human rotavirus isolated in Kunming in 2002 and 2003 were related to Wa, and their amino acids were highly conservative. The symptom of heavy diarrhea didn't seem to be associated with NSP4 genetic variations,but the precise mechanism of the correlation between the prevalence of the diarrhea and the rotavirus infection needed further investigation.
