rhKGF-2 Liniment Accelerate Healing of Burn Wound in Rabbit

摘  要:目的 研究rhKGF-2涂膜剂对皮肤深Ⅱ度烫伤的治疗作用。方法 用预热100℃的100g砝码在家兔背部脱毛部位烫8秒钟,形成4个直径约2.5cm的深Ⅱ度烫伤口,第2天开始每天伤口涂抹rhKGF-2涂膜剂150μl/伤口(30、15和7.5μg/伤口)1次,每4天描记伤口面积并计算伤口愈合率,烫伤后16天取伤口及其周围皮肤组织,石蜡包埋、切片、Masson’s三色法染色,MOTIC Advanced 3.2形态分析软件分析新生上皮细胞形成面积、厚度和迁移距离等。结果 rhKGF-2涂膜剂30μg/伤口和15州伤口对家兔皮肤深Ⅱ度烫伤的愈合均有明显的促进作用,能加快伤口愈合速度,促进上皮细胞再生,使新生上皮面积、平均厚度和移行距离明显增加。结论 rhKGF-2涂膜剂能明显加快皮肤烫伤后上皮细胞的增生与迁移,加快烫伤皮肤的愈合过程。Objective To observe the effect of recombinant human keratinocyte growth factor-2 (rhKGF-2) liniment on the healing of deep Ⅱ degree bum wound. Methods The back skin of each rabbit was burned for 8 seconds by 100℃ weights with diameter 2.5cm to become four deep Ⅱ degree burn wounds. The experimental animals were divided into 6 groups:the wound surfaces were daubed with saline solution as negative control group, liniment vehicle in vehicle group, rhKGF-2 solution in positive group, and rhKGF-2 liniments in the rest three groups (200, 100 and 50 μg/ml) daily for 15 days. The wound areas were measured at 4^th d, 8^th d, 12^th d and 16^th d. On the 16^th postinjury.Skin tissue biopsy specimens of the wound were taken. The area and thickness of neoepithelia tissue and the migrated distances of neoepidermal cells were detected with MOTIC Advanced 3.2 morphological analysis software after Masson's Trichrome staining. Results rhKGF-2 liniment accelerated deep Ⅱ degree burn wound in rabbit,accelerated the healing rate of wound, improved reepidermidation and increased the migrated distance, average area and thickness of neoepithelium. Conclusion These results showed that rhKGF-2 liniment could significantly accelerate wound healing in rabbit in a dose-dependently manner.
