摘 要:用磷酸苯二钠法测定新扬州鸡血浆碱性磷酸酶(PAKP)活性。结果表明,家系间、性别间和同功酶类型间有显著差异;随日龄增加,PAKP活性逐渐下降,开产前回升;产蛋后4~5h升至最高,之后,逐渐下降,20h降至最低;采食后,血浆碱性磷酸酶水平很高,6~18h降至较低水平,但相对稳定。聚丙稀酰胺凝胶电泳结果表明,该酶同功酶可分为F型(含有AKPF带)和S型(含有AKPs带)。通常情况下,F型酶和S型酶各具2条带,其相对含量随日龄变化明显。The plasma alkaline phosphatase (PAKP) levels were analysed by disodium phenyl orthophosphate method in new Yangzhou chickens. The results showed that difference among the families, sexes and isoenzyme types in the PAKP levels could be seen obviously. The levels of PAKP decreased gradually during the 6 ̄18 weeks of age, but increased before laying the first egg. Regular changes of PAKP levels were observed in the laying or hungry hens. It′s found that there were two zymograms through isoenzymes analysis with polyacrylamide gel electrophoresis, F type (AKP F band) and S type (AKP S band). In general, each type had 2 bands, and relative content changed obviously with days of age.
