摘  要:目的:观察绞股蓝总苷对老龄大鼠的抗氧化作用,探讨其延缓衰老的作用机制。 方法:实验于2004-03/2005-02在郑州大学医学院完成。选用健康老龄(18个月)SD雄性大鼠50只,随机分为5组,每组10只。空白对照组给予蒸馏水灌胃,1次/d;人参皂甙组给于人参皂甙20mg/kg灌胃,1次/d;绞股蓝总苷40,20及10mg/kg组,分别给于相应剂量绞股蓝总苷灌胃,1次/d。灌胃30d,取大鼠血清和脑组织,用硫代巴比妥酸法测定脂质过氧化物含量;用羟胺法检测超氧化物歧化酶活性及免疫器官指数。 结果:50只大鼠均进入结果分析。①绞股蓝对衰老大鼠胸腺指数和脾脏指数的影响:绞股蓝40mg/kg组和20mg/kg组胸腺指数和脾脏指数明显高于空白对照组[绞股蓝40mg/kg组(18.98±1.95),(22.12+2.41),20mg/kg组(17.98±1.64),(22.89±1.98),空白对照组(16.02+2.03),(18.79±2.37),P<0.05]。②绞股蓝对衰老大鼠血清及脑组织中超氧化 物歧化酶和丙二醛含量的影响:绞股蓝40mg/kg组和20mg/kg组血清超氧化物歧化酶活力显著提高;脑组织中超氧化物歧化酶活力亦显著高于空白对照组[(176.24±19.56),(154.35±17.23),(113.14±13.56)kNu/L,P<0.01];绞股蓝40mg/kg组大鼠血清丙二醛含量显著降低;绞股蓝40mg/kg组和20mg/kg组脑组织中丙二醛含量显著低于空白对照组[(102.24±11.32),(132.54±12.03)。(199.32±20.15)μmol/L,P<0.01)。 结论:绞股蓝总苷能增强老龄大鼠机体抗氧化能力,延缓机体衰老。AIM: To observe the antioxidation effect of gypenosides in aged rats and study its mechanism of delaying aging. METHODS: The experiment was done in the Medical College of Zhengzhou University from March 2004 to February 2005. Fifty male SD rats at 18 months of age were selected and randomized into five groups with 10 in each group: distilled water in blank control group (once a day), 20 mg/kg ginseng saponins in ginseng saponin group(once a day), 40, 20 and 10 mg/kg gypenosides in three treatment groups (once a day). Thirty days after treatment, both the activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in serum and brain were assayed, and immune organ index were determined. RESULTS: All the 50 rats were involved in the result analysis. ①Effect of gypenosides on thymus index and spleen index in aged rats: Thymus index and spleen index in the 40 mg/kg and 20 mg/kg gypenosides groups were higher than those in the control group[40 mg/kg group: 18.98±1.95, 22.12 ±2.41; 20 mg/kg group: 17.98±1.64,22.89±1,98; control group: 16.02±2.03, 18.79±2.37; P〈 0.05]. ②Effects of gypenosides on SOD activity and MDA content in serum and brain: The activity of SOD in serum was significantly increased in the 20 mg/kg and 40 mg/kg gypenosides groups, and the activity of SOD in brain was significantly higher than that in the control group [(176.24±19.56), (154.35±17.23), (113.14±13.56)kNu/L],P 〈 0.01], while the content of MDA in serum was decreased remarkably in the 40 mg/kg gypenoside group, and the MDA content in brain of the 20 mg/kg and 40 mg/kg groups was lower than that in the control group[(102.24 ±11,32), ( 132.54±12.03 ), ( 199.32±20.15)μmol/L,P 〈 0.01]. CONCLUSION: Gypenosides can strengthen the capability of anti-aging so as to delay aging in aged rats.
