紫芪方对IEC-6小肠上皮细胞缺氧复氧损伤后细胞凋亡及线粒体膜电位的影响

摘　　要：目的:观察缺氧复氧对IEC-6肠上皮细胞凋亡和线粒体膜电位的影响,探讨保护肠道屏障功能的复方中药——紫芪方对IEC-6 肠上皮细胞缺氧复氧损伤的保护作用及机制。方法:建立IEC-6肠上皮细胞缺氧复氧损伤模型。采用血清药理学方法,在细胞培养液中加入不同给药剂量(间隔1h,两次给药,20 g/kg)获取的紫芪方药物血清、参附药物血清及生理盐水血清(S)。应用流式细胞仪检测细胞凋亡率,采用激光共聚焦显微镜测定线粒体膜电位变化。结果:IEC-6肠上皮细胞缺氧复氧损伤后细胞凋亡率明显升高(P<0.01),线粒体膜电位明显降低(P<0.01);与损伤模型对照组比较,紫芪方药物血清能明显抑制IEC-6肠上皮细胞凋亡(P<0.05).并具有保护IEC-6 肠上皮细胞线粒体膜电位的作用(P<0.05)。结论:缺氧复氧可导致IEC-6肠上皮细胞凋亡数量明显升高和线粒体膜电位降低。紫芪方含药血清可减少细胞凋亡数量及保护细胞线粒体膜电位。紫芪方保护肠道的机制可能与抑制细胞异常凋亡有关。Objective： To explore the protective effect and mechanism of Ziqi- liquid, a preparation of Chinese herbal medicine for antagonizing two-hit, containing serum on hypoxia reoxygenation damaged intestinal epithelial cell（IEC-6）. Methods： Serving Ziqi-liquid drug＇s serum as treatment drug. Cultured IEC-6 cells were divided into four groups： control group（C）, hypoxia reoxygenation group（H/R）, SJ-2 group and SF group（two dose 20 g/kg interval 1 h）. After the cells in the four groups suffered 1 h of hypoxia followed by I h of reoxygenation. MMP was detected by laser scanning confocal microscopy and apoptosis and dead cell ratio were evaluated with Annexin-VFLOUS to see if there any difference among SJ-2, C. S and H/R group. Results： After hypoxia-reoxygenation, MMP in H/R group was significantly decreased and apoptosis and dead cell ratio in H/R group significantly increased compared with C group（ P 〈 0. 01） . These changes were significantly lessened by Ziqi-liquid containing serum. Conclusion： Ziqi-liquid containing serum have significant p tective effect on hypoxia reoxygenation induced injury, of intestinal epithelial cell in vitro. The probable mechanism may be concerned with the inhibition of cell apoptosis after hypoxia- reoxygenation.
