摘 要:目的:观察不同剂量的甘露消毒丸可否引起大鼠肾损害的发生。方法:SD大鼠32只随机分为4组:(1)空白对照组(C组,n=8):给以正常饲料饲养;(2)实验对照组(C2组,n=8):给以1.5%的低盐饲料饲养;(3)低剂量甘露消毒丸组(L组,n=8):给以低盐饮食及甘露消毒丸(7.5g·kg-1·d-1)饲养;(4)高剂量甘露消毒丸组(H组,n=8):给以低盐饮食及甘露消毒丸(15g·kg-1·d-1)饲养。16周实验结束时处死大鼠。分别进行尿蛋白定量、尿β2-MG、尿N-乙酰-β-D-葡萄糖苷酶(NAG)、肾功能、肾脏病理等方面的检查。结果:H组在用药16周后血肌酐、尿素氮、NAG均有明显升高(P<0.05);L组在用药16周血肌酐、尿素氮虽有升高,但与C2组相比,无统计学差异(P>0.05)。L组与H组在用药后16周尿蛋白定量及β2-MG与C2组相比均无统计学差异(P>0.05)。肾脏病理检查可见H组动物在16周时有肾小管上皮细胞空泡变性,部分崩解脱落,管腔扩张;L组动物肾脏病理有轻微改变。结论:本研究应用高剂量的甘露消毒丸同时合并应用低盐饮食,观察到16周时诱导了大鼠肾小管损害的发生及血肌酐、尿素氮和NAG的升高。但是诱导实验动物发生慢性肾间质纤维化的时间及条件尚待进一步探讨。Objective:To observe whether different doses of Ganluxiaodu pellet can induce renal injury in SD rats. Methods:32 male SD rats (220-250 g) were randomly divided into four groups. (1)normal group fed with normal diet (C group,n =8) ;(2)experiment control group fed with low salt, normal-protein diet (1.5% sodium) (C2 group, n =8); (3)low doseg treated group treated with low salt, normal - protein diet ( 1.5% sodium) and low dose Ganluxiaoduwan (7.5 g·kg^-1·d^-) (L group, n =8);(4)high doseage group treated with low- salt, normal- protein diet (1.5% sodium) and high dose Ganluxiaoduwan( 15 g·kg^-1·d^- ) (H group, n = 8). All rats were sacrificed on 16th week. Quantitative analysis of urine β2 microglobulin (β2-MG), N - acetyl beta D- glucosarnidase (NAG), and renal function t as well as pathological study were performed respectively. Results. The blood urea nitrogen (BUN) levels, serum creatinine (Scr) levds and the NAG ldvds in urine were all significantly increased in H group ( P〈 0.05) afer 16 weeks treatment, and there were little increase in BUN, Scr and NAG in L group. But the 24 hours urine protein and 132 - MG were not increased markedly in both L and H group(P 〉0.05). The renal pathologic examination showed normal glomerular structure and disappearance of renal tubular epithelial in H group, and there were little change in L group. Conclusion: In this study we found that low salt, normal - protein diet ( 1.5 % sodium) combied with high dose Ganluxiaoduwan( 15 g·kg^-1·d^- ) may induce renal tubular injury and elevation of BUN, Scr and NAG in rats. Further studies should be carried out to make sure the time and conditions needed to induce the chronic renal interstitial fibrosis in experiment animal model.
